摘要
目的研究电针夹脊穴对退变椎间盘内感觉神经相应DRG Netrin-1的表达的影响。为临床治疗本病提供理论依据。方法20只兔分为正常组、模型组、假模型组、模型+电针组、模型+假电针组5组,每组4只,模型组采用轴向加压方法建立椎间盘退变模型,模型+电针组在造模后采用电针治疗相应夹脊穴28 d。实验结束后取出相应DRG(L),采用Western blot和免疫组化方法处理标本。结果Western blot和免疫组化均发现:Netrin-1在正常组及假模型组L节段DRG中表达丰富;模型组DRG的Netrin-1和mRNA表达下降,电针28 d后,Netrin-1及mRNA的表达水平上调。结论DRG中Netrin-1表达下调可能是引起退变椎间盘内感觉神经内向生长的原因,电针治疗相应夹脊穴可以通过逆转这一现象从而延缓椎间盘退变。
Objective To study the effect of electroacupuncture at Jiaji acupoints on the expression of dorsal root ganglion(DRG)Netrin-1 in sensory nerves in degenerated intervertebral discs to provide theoretical basis for the clinical treatment of this disease.Methods Twenty rabbits were divided into 5 groups:normal group,model group,sham model group,model+electroacupuncture group,model+sham electroacupuncture group,4 rabbits in each group,and the model group used axial compression to establish a model of intervertebral disc degeneration in the model+electroacupuncture group.The corresponding Jiaji acupoints were treated with electroacupuncture for 28 days after modeling.After the experiment,the corresponding DRG(L)was taken out,and the specimens were processed by Western blot and immunohistochemistry.Results Western blot and immunohistochemistry both found that Netrin-1 was abundantly expressed in DRG in the Lsegment of the normal group and the sham model group.The Netrin-1 and mRNA expression of DRG in the model group decreased.After 28 days of electroacupuncture,Netrin-1 and the expression level of mRNA was up-regulated.Conclusion The down-regulation of Netrin-1 expression in DRG may be responsible for the inward growth of sensory nerves in degenerated intervertebral discs.Electroacupuncture treatment of the corresponding Jiaji acupoints can reverse this phenomenon and delay the degeneration of intervertebral discs.
