摘要
目的:建立HPLC法和UV法分别测定新疆两色金鸡菊中绿原酸和总黄酮的含量。方法:采用HPLC法测定绿原酸含量,Agilent Eclipse Plus C18柱(250 mm×4.6 mm,5μm),以流动相0.1%磷酸水溶液-乙腈,梯度洗脱,流速1.0 mL/min,检测波长348 nm,柱温30℃。采用UV法测定总黄酮含量,NaNO2-Al(NO3)3-NaOH方法显色,以芦丁为对照,在波长522 nm处检测。结果:方法在考察的浓度范围内线性关系良好,精密度、准确度和稳定性RSD均小于5%。不同产地新疆两色金鸡菊绿原酸含量为1.39%~5.76%,总黄酮含量为19.10%~28.40%。结论:本试验建立了含量测定的方法,简便可靠,重现性好,适用于两色金鸡菊中绿原酸和总黄酮的含量测定。
Objective:To develop HPLC and UV methods for determination of chlorogenic acid and total flavonoids in Coreopsis tinctoria Nutt.Methods:HPLC analysis was performed on an Agilent Eclipse Plus C18(250 mm×4.6 mm,5μm).The mobile phase was acetonitrile and 0.1%phosphoric acid aqueous solution with gradient elution.The flow rate was 1 mL/min,the detection wavelength was 348 nm and the column temperature was 30℃.The content of total flavonoids in Coreopsis tinctoria Nutt was detected by NaNO2-Al(NO3)3-NaOH colorimetry at a wavelength of 522 nm.Results:Satisfactory linearity was achieved in the linear range.The RSD of precision,accuracy and stability were less than 5%.The content of chlorogenic acid ranges from 1.39%to 5.76%and total flavonoids ranges from 19.10%~28.40%.Conclusion:The assay method established in this experiment meet the requirements of methodological verification.The method is simple,reliable and reproducible.It could be used for determination of chlorogenic acid and total flavonoids in Coreopsis tinctoria Nutt.
