摘要
目的从血液流变学和血小板活化探讨补阳还五汤改善气虚血瘀模型大鼠血瘀证的作用机制。方法40只SD大鼠,随机分为空白组、气虚血瘀模型组、补阳还五汤高剂量组(14.0 g·kg^-1)、补阳还五汤低剂量组(3.5 g·kg^-1),每组10只。大鼠以5%水合氯醛腹腔注射麻醉,沿颈部正中线做1 cm切口,一次性向气管内注射博莱霉素生理盐水溶液(5 mL·kg^-1)。注射完后立即将大鼠直立旋转,使药液在肺内充分分布均匀,缝合创口。造模后第2天开始腹腔注射给药。结果与模型组比较,补阳还五汤能够显著降低全血黏度(P<0.05)、血浆黏度(P<0.05),可显著升高6-keto-PGF1α、cAMP含量(P<0.05),显著降低TXB2、β-TG含量(P<0.05);补阳还五汤高剂量组可显著降低cGMP、vWF、Fn含量。结论补阳还五汤能降低气虚血瘀模型大鼠全血黏度、血浆黏度,改善血液的高黏状态。补阳还五汤通过改善血小板聚集、黏附,抑制血小板释放亢进的状态等方面来缓解气虚血瘀模型大鼠病理状态。
Objective To explore the mechanism of Buyang Huanwu Decoction in improving blood stasis syndrome in rats with Qi deficiency and blood stasis by blood rheology and platelet activation.Methods 40 SD rats were randomly divided into blank group,Qi deficiency and blood stasis model group,Buyang Huanwu high dose group(14.0 g·kg^-1)and Buyang Huanwu decoction low dose group(3.5 g·kg^-1),10 per group.The rats were anesthetized by intraperitoneal injection of 5%chloral hydrate,and 1 cm incision was made along the midline of the neck.The bleomycin physiological saline solution(5 mL·kg^-1)was injected into the trachea at one time.Immediately after the injection,the rats were rotated upright,so that the drug solution was well distributed in the lungs and the wound was sutured.Intraperitoneal administration was started on the second day after modeling.Results Compared with the model group,Buyang Huanwu Decoction significantly reduced whole blood viscosity(P<0.05),plasma viscosity(P<0.05),and significantly increased 6-keto-PGF1αand cAMP content(P<0.05).The content of TXB2 andβ-TG was significantly decreased(P<0.05).The high dose of Buyang Huanwu Decoction could significantly reduce the contents of cGMP,vWF and Fn.Conclusion Buyang Huanwu Decoction can reduce the blood viscosity and plasma viscosity of rats with Qi deficiency and blood stasis,and improve the high viscosity of blood.Buyang Huanwu Decoction can relieve the pathological state of rats with Qi deficiency and blood stasis by improving platelet aggregation,adhesion,and inhibiting the state of platelet hyperthyroidism.
