摘要
目的采用网络药理学方法研究穿心莲治疗肺炎的作用机制。方法利用中药系统药理学数据库与分析平台(TCMSP)对穿心莲的成分及靶点进行收集,通过比较毒物基因组学数据库(CTD)对肺炎的疾病靶点进行收集,并把收集到的药物靶点与疾病靶点进行匹配;使用Cytoscape建立"药物成分-靶点"网络图和靶点蛋白网络图;基于ClueGO对核心靶点进行GO功能富集和KEGG通路富集分析;最后,把候选靶点标注于最相关的通路上进行分析。结果通过TCMSP及CTD得到穿心莲治疗肺炎的24种药物成分及71个候选靶点,其中核心靶点45个;GO功能富集分析结果:分子功能(MF)54个、生物学过程(BP)295个及细胞组分(CC)5个,KEGG通路富集分析与筛选后共获得相关通路36条。结论初步分析与预测穿心莲"多成分-多靶点-多通路"治疗肺炎的分子作用机制,为进一步开展其药物成分开发及机制研究提供科学依据。
Objective To analyze Chuanxinlian(Andrographis Herba)in the treatment of pneumonia based on network pharmacology.Methods The ingredients of herb and their corresponding targets were collected from traditional Chinese medicine system pharmacology database(TCMSP),the targets related to pneumonia were collected from Comparative Toxicogenomics Database(CTD),and match the collected herb targets with disease targets;Herb ingredients-targets network and protein interacting network were built by Cytoscape software;ClueGO was used to enrich the gene ontology and pathway;Finally,the candidate targets were mapped to the graph of the most relevant pathway.Results The 24 herb ingredients and 71 candidate targets were screened out from TCMSP and CTD,and including 45 core targets;The items of GO function enrichment analysis:molecular function 54 items,biological process 295 items,cellular component 5 items,a total of 36 related pathways were obtained from KEGG pathway analysis.Conclusion The results of this study preliminarily analysised and predicted the molecular mechanism of Chuanxinlian(Andrographis Herba)treating pneumonia through"multi-ingredients,multi-targets,multi-pathways",and provide a scientific basis for further experimental research on its ingredients and mechanism research.
