膀胱癌膜蛋白相关预后模型的构建与验证

Construction and validation of a membrane protein-related prognostic model for bladder cancer

摘要 目的探讨膜蛋白(membrane proteins,MP)在膀胱癌(bladder cancer,BLCA)预后中的作用,并建立一种基于MP预测BLCA患者生存的预后模型。方法从癌症基因组图谱(the cancer genome atlas,TCGA)数据库下载BLCA的MP序列数据,测定肿瘤组织和正常组织MP的差异表达并进行功能富集分析。通过单变量和多变量Cox回归分析鉴定独立预后相关MP,构建风险评分模型。绘制Kaplan-Meier和受试者工作特征(receiver operating charac teristic,ROC)曲线来评估该预后模型的性能。建立列线图,验证其预后价值和关键MP的表达。结果在筛选出的395个蛋白中,有243个上调,152个下调。根据单因素及多因素生存分析发现三个独立的预后相关蛋白(PCDHGA2、ITGB7、FLRT2),并构建预后模型。根据预后模型,高危亚组患者总生存期低于低危亚组患者,ROC曲线下面积为0.725和0.709。对TCGA数据库的数据进行分析,进一步验证了预后分析模型,建立了线型图,MP对BLCA的预后有良好的预测价值。结论开发并验证了BLCA良好的预后模型,该模型可能有助于开发BLCA患者预后的新生物标志物。 Objective To investigate the role of membrane proteins(MPs)in bladder cancer(BLCA)prognosis and to develop a prognostic model for survival of BLCA patients based on MPs.Method MP sequence data for BLCA were downloaded from the cancer genome atlas(TCGA)database,and the differential expression of MPs between tumor and normal tissues was determined and analyzed for functional enrichment.Univariate and multivariate Cox regression analyses were performed to identify independent prognosis-related MPs and to construct risk score models.Kaplan-Meier and ROC curves were plotted to assess the prognostic model’s performance.Column line plots were generated to validate its prognostic value and expression of key MPs.Result Of the 395 proteins screened,243 were upregulated and 152 were down regulated.Three independent prognosis-related proteins(PCDHGA2,ITGB7,and FLRT2)were identified based on univariate and multifactorial survival analyses,and prognostic models were constructed.According to the prognostic model,the overall patient survival in the high-risk subgroup was lower than that of patients in the low-risk subgroup,and the areas under the ROC curve were0.725 and 0.709,respectively.The data from the TCGA database were analyzed to further validate the prognostic analysis model and establish a line graph.The MP had a good predictive value for the BCLA prognosis.Conclusion A prognostic model for BLCA was developed and validated,which may help to identify new biomarkers for BLCA patient prognosis.
