摘要
目的:探讨血浆外泌体miR-1260b在肺腺癌中的表达及其与临床特征的关系。方法:收集并提取肺腺癌患者血浆中外泌体,通过透射电镜(transmission electron microscopy,TEM)、粒径分析(nanoparticle tracking analysis,NTA)、Western blot鉴定外泌体的大小、分布情况。采用qRT-PCR检测外泌体miR-1260b的表达。结果:外泌体广泛存在于肺腺癌患者血浆中。亚组分析发现,与肿瘤≤3 cm组相比,血浆外泌体miR-1260b在肿瘤>3 cm组中呈高表达(P=0.010);与N0组相比,血浆外泌体miR-1260b在N1(P<0.05)、N2(P<0.001)组中呈高表达;与非气道播散组相比,血浆外泌体miR-1260b在气道播散组中高表达(P<0.001)。与未复发/转移组相比,血浆外泌体miR-1260b在复发/转移组中高表达(P=0.038)。结论:血浆外泌体miR-1260b在肺腺癌中高表达,且与肿瘤大小、气道播散、复发/转移相关,可作为肺腺癌患者诊治的分子靶标。
Objective:This study aimgs to investigate the expression of plasma exosomal miR-1260 b in patients with lung adenocarcinoma and its correlation with clinical features.Methods:Exosomes were extracted from the plasma of the patients with lung adenocarcinoma.The size and distribution of exosomes were identified by transmission electron microscopy(TEM),nanoparticle tracking analysis(NTA)and Western blot.qRT-PCR was used to detect the expression of exosomal miR-1260 b.Results:Exosomes wre widely present in the plasma of patients with lung adenocarcinoma.Subgroup analysis found that plasma exosomal miR-1260 b was highly expressed in>3 cm compared with≤3 cm group(P=0.010);Compared with the N0 group,plasma exosomal miR-1260 b was increased in the N1(P<0.05)and N2(P<0.001)groups;Compared with the spread without air spaces group,plasma exosomal miR-1260 b was elevated in the spread through air spaces group(P<0.001).Compared with the non-relapse/metastasis group,plasma exosomal miR-1260 b was highly expressed in the relapse/metastasis group(P=0.038).Conclusion:Plasma exosomal miR-1260 b is highly expressed in patients with lung adenocarcinoma,and is related to tumor size,stas and recurrence/metastasis.It can be used as a molecular target for the diagnosis and treatment of lung adenocarcinoma patients.
