摘要
目的:探讨孙思邈治疗消渴病的遣方用药规律。方法:整理《千金要方》卷二十一、《千金翼方》卷十九中治疗消渴病的76首方剂用药,将其录入仓公诊籍国医脉案数据记录挖掘系统统计药物使用、性味归经频次,采用关联分析、聚类分析法挖掘孙思邈治疗消渴病的用药特点和配伍规律,并通过生物信息学分析工具BATMAN-TCM对聚类分析所得核心方药进行GO富集分析和KEGG分子信号通路富集分析,预测其治疗糖尿病的可能分子作用靶点。结果:共纳入方剂76首,所用药物136味。药物使用频次前5位分别为天花粉、甘草、麦门冬、黄连、生地黄;频次前3位的药类分别为清热泻火药、补气药、补阴药;药性使用前4位分别为平、寒、微寒、温,药味使用频次前4位分别为甘、苦、辛、微苦;前5位归经依次是胃经、肺经、心经、脾经、肾经;关联分析得到常用2味、3味药物组合共计30个;聚类分析得到3类常用方药组合;GO富集分析共获得8个分子功能、8个细胞组分、20个生物过程,KEGG分子信号通路富集分析得到28条细胞信号通路。结论:孙思邈治疗消渴病用药以清热滋阴,补气益肾为主,寒温并用、阴阳互补,核心方药治疗糖尿病是多靶点、多通路的复杂机制,其机制与调节细胞内能量代谢、抑制炎症因子、调节肠道菌群等相关。
Objective:To explore the regularity of Sun Simiao’s prescription in treating diabetes.Methods:Sorted out 76 prescriptions for diabetes in Volume 21 of"Qian Jin Yao Fang"and Volume 19 of"Qian Jin Yi Fang",and recorded them into the data mining system of Cang Gong’s national medical pulse records.Statistics on drug use and frequency of sexual and flavor tropism were made,and Sun Simiao’s drug use characteristics and compatibility rules for diabetes were discovered by using correlation analysis and cluster analysis,The core prescriptions obtained from cluster analysis were analyzed by GO enrichment analysis and KEGG molecular signal pathway en-richment analysis through the bioinformatics analysis tool BATMAN-TCM to predict their possible molecular targets for the treatment of diabetes.Results:A total of 76 prescriptions were included and 136 of them were used.The first 5 cases of drug use were trichosanthin,licorice,Ophiopogon,Rhizoma Coptidis,and Rehmannia;the first 3 of the frequency of the frequency were heat clearing and diarrhea,supplementing qi and nourishing Yin respectively.Before the use of the drug,the 4 were flat,cold,cold and warm,and the first 4 were sweet,bitter,symplectic and bitter,respectively,and the first 5 were the following:Gastric meridian,lung meridian,heart meridian,splenic meridian,kidney meridian;correlation analysis obtained 30 commonly used 2 flavors and 3 flavors;3 kinds of common prescrip-tions were obtained by cluster analysis;8 molecular functions,8 cell components and 20 biological processes were obtained by GO en-richment analysis,and 28 cell signaling pathways were obtained by Tong Lufu set analysis.Conclusion:The treatment of Sun Simiao for the treatment of diabetes is to clearing heat and nourishing Yin,nourishing qi and nourishing kidney,combining cold and warm with Yin and Yang.The core prescription is a complex mechanism of multi-target and multi pathway in the treatment of diabetes.Its mechanism regulates intracellular energy metabolism,inhibits inflammatory factors and regulates intestinal microflora.
