摘要
目的研究纳米二氧化钛暴露对子代大鼠的空间学习记忆行为的影响。方法90只SD大鼠分为纳米二氧化钛实验组和对照组,每组30只雌鼠、15只雄鼠。实验组灌胃1次/d给予250 mg/kg·bw的纳米二氧化钛混悬液,对照组给予不加纳米二氧化钛粉末的溶媒。灌胃2周后进行交配,亲代雄鼠连续灌胃染毒共10周,雌鼠在交配期、妊娠期和哺乳期持续灌胃染毒至处死。子代大鼠从离乳持续灌胃染毒至处死。期间每周1次进行子鼠体重观察,进行Morris水迷宫实验(隐匿平台测试训练、空间探索实验和可视平台实验)对空间学习记忆能力评价。实验结束处死后进行脏器称重并计算脏器系数,行病理组织学检查。结果与对照组比较,实验组子代雌雄大鼠在出生后42和56 d的体重降低,差异有统计学意义(P<0.05,P<0.01)。Morris水迷宫实验中,对照组和实验组结果差异无统计学意义。实验组子代雌雄大鼠脑绝对重量和脑/体比值与对照组相比,差异无统计学意义(P>0.05),组织病理学检查未发现异常。结论在本实验研究条件下,粒径为25~40 nm的纳米二氧化钛在一代繁殖毒性扩展试验(OECD 443)中,未观察到对子代大鼠空间学习记忆有影响。
Objective To explore the effect of exposure to nano-TiO2 on the spatial learning and memory ability of offspring rats.Methods Ninety SD rats were divided into nano-TiO2 exposed group and control group,30 females and 15 males in each group.Exposed group was given nano-Ti O2 by intragastrical administration at the doses of 250 mg/kg·bw,and the control group was given equal volume solution without nano-TiO2.The exposure was continued for 2 weeks before mating.Male rats were exposed continuously for ten weeks.Female rats were exposed throughout pregnancy and lactation.Offspring were perfused nano-Ti O2 into stomach from weaning to execution.The weight of offspring was observed once a week.Morris water maze(recessive platform training,space exploration experiment,visible platform experiment)was used to evaluate spatial learning and memory ability.The organs were weighed and performed by histopathological examination.The organ coefficients were also calculated.Results Compared with the control group,the weight of the male and female rats in the experimental group decreased significantly at postnatal day 42 and 56(P<0.05,P<0.01).There was no significant difference in Morris water maze test.Compared with the control group,the weight and organ coefficients of brain in experimental group were not significantly different(P>0.05).And no abnormalities were found in histopathological examination.Conclusion In the one-generation reproduction toxicity expansion test(OECD 443),the nano-TiO2 with a particle size of 25~40 nm was not observed to affect the spatial learning and memory behavior of the offspring rats.
