SOCS-3及HMGB1表达水平与过敏性紫癜性肾炎发生发展的关系 被引量:4

The relation between SOCS-3,HMGB1 and the occurrence and development of rat model with purpura nephritis

摘要 目的探讨细胞因子信号转导抑制蛋白3(SOCS-3)、高迁移率族蛋白1(HMGB1)表达水平与过敏性紫癜性肾炎(HSPN)发生发展的关系。方法将24只SPF级SD大鼠随机分为对照组、紫癜组和紫癜肾组,每组8只。构建HSPN大鼠模型成功后,取大鼠全血、尿液及肾脏组织分别进行HE染色、尿蛋白、IgA水平检测,并检测肾脏组织中SOCS-3、HMGB1的蛋白表达水平。结果HE染色显示,紫癜肾组大鼠肾间质血管明显扩张充血,肾小球出血,出血量+++,表示造模成功。紫癜肾组尿蛋白含量低于紫癜组和对照组[(0.133±0.006)μg/mL vs.(0.226±0.005)μg/mL vs.(0.149±0.005)μg/mL],差异有统计学意义(P均<0.001)。紫癜肾组大鼠肿瘤坏死因子-α、白介素-6水平均显著高于紫癜组和对照组,差异有统计学意义(P<0.001)。紫癜肾组HMGB1与SOCS-3蛋白表达量显著高于对照组[HMGB1:(25874±2208)vs.(866±66),SOCS-3:(27471±4901)vs.(1540±1088),差异有统计学意义(P<0.001),但与紫癜组比较,差异无统计学意义(P>0.05)。紫癜肾组肾脏组织IgA蛋白沉降水平显著高于对照组(101.300±8.189)vs.(64.000±4.697),差异有统计学意义(P<0.001),但与紫癜组比较,差异无统计学意义(P>0.05)。结论与紫癜大鼠模型比较,SOCS-3、HMGB1在紫癜肾大鼠模型中明显升高。 Objective To investigate the relationship of suppressor of cytokine signaling-3(SOCS-3)and high mobility group protein 1(HMGB1)with the development and progression of henoch-schonlein purpura nephritis(HSPN).Methods The SD rats were collected and randomly divided into three groups,which were comtrol group,henoch-schonlein purpura(HSP)group and HSPN group.Each group had 8 rats.HSPN rat model was constructed.Whole blood,urine,and kidney tissues of rats were sampled for pathology,urine protein,and Ig A level tests,and the protein expression levels of SOCS-3and HMGB1 in kidney tissues were also measured.Statistical analysis of test data was conducted.Results HE staining results showed that the renal interstitial vessels of rats in the HSPN group were significantly dilated and congested,with glomerular hemorrhage(bleeding volume+++),indicating that the model was successfully established.Urinary protein BCA results showed that the urinary protein level of rats in the HSPN group was significantly lower than that in the HSP and control groups[(0.133±0.006)μg/m L vs.(0.226±0.005)μg/m L vs.(0.149±0.005)μg/m L](P<0.001).ELISA results showed that tumor necrosis factor-α,interleukin 6 levels of rats in the HSPN group were significantly higher than those in the HSP and control groups(P<0.001).Immunohistochemical results showed that protein expression levels of HMGB1 and SOCS-3 of rats in the HSPN group were significantly higher than those in the control group[HMGB1:(25874±2208)vs.(866±66),SOCS-3:(27471±4901)vs.(1540±1088)(P<0.001),but were no different than HSP group(P>0.05).Immunofluorescence results showed that the Ig A protein sedimentation level of rats in the HSPN group was significantly higher than that in the control group(101.300±8.189)vs.(64.000±4.697)(P<0.05),but were no different than HSP group(P>0.05).Conclusion The protein expression levels of SOCS-3 and HMGB1 were significantly increased in the HSPN group in comparison with the control group.
