摘要
目的探讨长链非编码RNA(lncRNA)和信使RNA(mRNA)在乳腺癌中的差异表达情况。方法组织样本采集于2018年9-11月,来自于新疆医科大学附属肿瘤医院。选取5例经病理诊断结果确认为乳腺癌的肿瘤组织及5例经病理证实无肿瘤侵犯的乳腺癌旁正常组织(癌旁组织)。采用博奥晶典长链非编码RNA芯片V4检测乳腺癌组织和癌旁组织lncRNA和mRNA的差异表达。结果相对于癌旁组织,乳腺癌组织中差异表达上调的lncRNA有551个,下调的lncRNA有959个,乳腺癌组织中差异表达前5位的lncRNA分别是XR_159043.1、TCONS_00013634、uc003gdg.1、TCONS_00009203、TCONS_00016441。在mRNA表达谱芯片中,相对于癌旁组织,乳腺癌差异表达上调的mRNA有529个,下调的mRNA有1129个;乳腺癌组织中差异表达前5位的mRNA分别是LEP、PLIN1、ADIPOQ、PRSS56、HEPACAM。根据所得差异表达的mRNA,通过基因本体论(GO)的数据库,疾病的富集分析可以观察到,富集到的疾病包括免疫系统疾病、获得性免疫缺陷综合征(AIDS)、肿瘤、中枢神经系统肿瘤等。富集到的京都基因与基因组百科全书(KEGG)通路包括磷酸化腺苷酸活化蛋白激酶(AMPK)信号通路、磷脂酰肌醇3激酶/蛋白激酶B(PI3K-Akt)信号通路、非受体酪氨酸激酶-信号转导及激活转录因子(Jak-STAT)信号通路、Ras信号通路、叉头框蛋白O(FoxO)信号通路。结论通过lncRNA芯片技术筛选出与乳腺癌相关的lncRNA与mRNA,为下一步探究lncRNA和mRNA在乳腺癌中的作用机制奠定了研究基础。
Objective To investigate the differential expression of long-chain non-coding RNA(lncRNA)and messager RNA(mRNA)in breast cancer.Methods Tissue samples were collected from September to November 2018,which were from the Affiliated Tumor Hospital of Xinjiang Medical University.Five tumor tissues confirmed by pathological diagnosis as breast cancer and five normal adjacent breast cancer tissues(adjacent tissues)confirmed by pathological examination without tumor invasion were selected.CapitalBio Technology Human LncRNA Array V4 designed with four identical arrays per slide(4×180 K format)was used to detect the differential expression of lncRNA and mRNA in breast cancer tissues and adjacent tissues.Results Compared with the adjacent tissues,there were 551 differentially up-regulated and 959down-regulated lncRNAs in breast cancer tissues.The top 5 lncRNA differentially expressed in breast cancer tissues were XR_159043.1,TCONS_00013634,uc003gdg.1,TCONS_00009203 and TCONS_00016441.In the mRNA expression profile chip,compared with the adjacent tissues,there were 529 differentially up-regulated mRNA and 1129 down-regulated mRNA in breast cancer.The top 5 mRNA differentially expressed in breast cancer tissues were LEP,PLIN1,ADIPOQ,PRSS56 and HEPACAM.Gene ontology(GO)enrichment analysis showed diseases observed including immune system diseases,acquired immune deficiency syndrome(AIDS),tumors,and central nervous system tumors.The enriched Kyoto Encyclopedia of Genes and Genomes(KEEG)pathway includes adenosine 5’-monophosphate-activated protein kinase(AMPK)signal pathway,phosphatidylinositol 3 kinase/protein kinase B(PI3K-Akt)signal pathway,just another kinase-signal transducers and activators of transcription(Jak-STAT)signal pathway,Ras signal pathway,and FoxO signal pathway.Conclusion This study screened lncRNA and mRNA related to breast cancer by chip technology,which laid the foundation for further research on the mechanism of lncRNA and mRNA in breast cancer.
