摘要
目的:研究湿热外环境在岭南湿热证炎症反应中的作用机理及茵陈蒿的干预机制。方法:实验一,将48只SPF级BALB/c小鼠随机分为4组:对照组[T(25±2)℃,HR 55%]、岭南湿热外环境模型组[T(33±2)℃,HR 85%]、高热组[T(33±2)℃,HR 55%]、湿阻组[T(25±2)℃,HR 85%],12 h暴露,造模28 d,每天记录小鼠进食量,每周记录小鼠体质量。分别于造模后第14天与第28天,采用酶联免疫吸附法(ELISA)检测各组小鼠白细胞介素-6(IL-6)、IL-18、IL-1β、肿瘤坏死因子-α(TNF-α)、核转录因子κB(NF-κB)及Toll样受体4(TLR4)等炎症因子血清水平。实验二,将50只SPF级BALB/c小鼠随机分为:正常组、岭南湿热外环境模型组、茵陈蒿低、中、高剂量组,将模型组与茵陈蒿3个剂量组放入人工气候箱[T(33±2)℃,HR 85%],12 h暴露,制造湿热外环境。造模后14 d,茵陈蒿3组小鼠连续7 d用3种不同浓度的茵陈蒿水煎剂灌胃。检测方法及指标同上。结果:实验一,造模后14 d、造模后14~28 d、造模后28 d,与对照组、高热组、湿阻组相比,模型组小鼠血清TNF-α含量增加(P<0.05,P<0.01)。各组IL-18、IL-6、NF-κB、TLR4比较,差异无统计学意义(P>0.05)。14~28 d,高热组IL-1β与岭南湿热外环境模型组比较,差异有统计学意义(P<0.01)。实验二,与模型组相比,茵陈蒿低、中、高剂量组血清TNF-α含量均降低(P<0.05,P<0.01)。各组IL-18、IL-1β、IL-6、NF-κB、TLR4比较,差异无统计学意义(P>0.05)。结论:湿热外环境通过上调血清TNF-α水平触发岭南湿热证炎症反应的机制是由“外湿之邪”与“外热之邪”共同作用所致,而茵陈蒿可通过干预这一机制改善此病理过程。
Objective:To explore the mechanism of the damp-heat external environment in the inflammatory response of Lingnan damp-heat syndrome and the intervention mechanism of Artemisiae Scopariae Herba.Methods:In experiment 1,48 SPF grade male BALB/c mice were randomly divided into four groups:the control group[T(25±2)℃,HR 55%],the Lingnan damp-heat external environment model group[T(33±2)℃,HR 85%],the high fever group[T(33±2)℃,HR 55%],and the dampness obstruction group[T(25±2)℃,HR 85%].After 12 hours of exposure,the model was established for 28 days.The food intake of the mice was recorded daily,and the body mass of the mice was recorded weekly.On the day 14 and 28 of modeling,enzyme-linked immunosorbent assay(ELISA)was used to detect serum levels of inflammatory factors such as interleukin-6(IL-6),IL-18,and IL-1β,tumor necrosis factor-α(TNF-α),nuclear transcription factorκB(NF-κB)and Toll-like receptor 4(TLR4)in each group of mice.In experiment 2,50 SPF grade male BALB/c mice were randomly divided into three groups:normal group:Lingnan damp-heat external environment model group,low-,medium-,and high-dose groups of Artemisiae Scopariae Herba.The model group and three groups of Artemisiae Scopariae Herba were placed in an artificial climate box[T(33±2)℃,HR 85%],and exposed for 12 hours to create a dampheat external environment.After 14 days of modeling,three groups of mice with Artemisiae Scopariae Herba were given intragastric administration of three different concentrations of Artemisiae Scopariae Herba decoction for 7 consecutive days.The detection method and indicators are the same as above.Results:In experiment 1,after 14 days,14-28 days and 28 days of modeling,compared with the control group,high fever group,and dampness obstruction group,the serum TNF-αof the model group mice was increased(P<0.05,P<0.01).There was no statistically significant difference in the comparison of IL-18,IL-6,NF-κB and TLR4 in each group(P>0.05).At days 14-28,IL-1βin high fever group compared with the Lingnan clamp-heat external environment model group,the difference was statistically significant(P<0.01).Experiment 2,compared with the model group,the serum TNF-αlevels in the low-,medium-,and high-dose groups of Artemisiae Scopariae Herba were decreased(P<0.05,P<0.01).There was no statistically significant difference in comparison of IL-18,IL-1β,IL-6,NF-κB and TLR4 in each group(P>0.05).Conclusion:The damp-heat external environment up-regulates serum TNF-αlevels to trigger the inflammatory response of Lingnan damp heat syndrome is caused by the joint action of"pathogenic factors of external dampness"and"pathogenic factors of external heat",and Artemisiae Scopariae Herba can improve this pathological process through this mechanism.
