摘要
单细胞转录组测序技术为表征细胞异质性,解析复杂的生物系统提供了强大的工具;生物医学研究的进一步发展不断地向该技术提出新的需求。经过过去十年的飞速发展,该技术的灵敏度在逐渐提高,并且通量发生了巨大的飞跃,也实现了全转录组和全长转录本测序。本文主要介绍单细胞测序技术的发展,在灵敏度提升,通量提高和实现全转录组测序和全长转录本测序上面临的技术挑战。我们也提出未来需要开发更优的单细胞转录组测序技术,开发自动化单细胞测序仪器,并需要将该技术与空间组学和多组学测序技术更好的结合。
Single-cell transcriptome sequencing technology provides a powerful tool for characterizing cell heterogeneity and analyzing complex biological systems.The further development of biomedical research continuously puts forward new challenges for this technology.After a decade of rapid development,the sensitivity of this technology is gradually improved,and the throughput has taken a huge leap.Full transcriptome and full transcriptome sequencing have also been achieved.This review introduces the development of single-cell transcriptome sequencing technology,and the technical challenges faced in improving sensitivity,increasing throughput,and achieving full-length transcript sequencing and whole transcriptome sequencing.We also proposed the need to develop advanced single-cell transcriptome sequencing technology in the future,develop automated single-cell sequencing instruments,and better integrate this technology with spatial omics and multi-omics sequencing technologies.
