摘要
萝卜硫素(SFN)是蔬菜中发现的抗癌效果最好的植物活性物质。采用DFT方法在B3YLYP/6-31+G水平下计算萝卜硫素及其衍生物的羰基氧5-O的ESP电荷、NBO电荷、HIR电荷和Mulliken电荷,通过用不同的基团单取代得到17种不同衍生物,利用Chem3D软件模拟了17种不同取代基的萝卜硫素衍生物的沸点,并通过拟合发现羰基氧5-O的NBO电荷值的相关系数值较大(R2=0.951,y=1 028.24+1 080.58x),利用信息理论指数拟合的羰基氧5-O的SGBP值的相关系数值最大(y=25.13+1.60x,R2=0.980)。还模拟了萝卜硫素及其-F代衍生物的质谱,对其谱图中的特征峰进行指认和归属,这对萝卜硫素及其衍生物的酶法合成提供了理论支持,也为材料工程与制药专业拓展实验识别其萝卜硫素衍生物以及构效关系提供了基础数据。
Sulforaphane(SFN) is the most potent anticancer plant active substance found in vegetables. The ESP charge, NBO charge, HIR charge and Mullliken charge of carbonyl oxygen 5-O of sulforaphane and its derivatives were calculated by DFT method at B3 YLYP/6-31+G level. Seventeen different derivatives were obtained by single substitution of different groups. The boiling points of 17 different substituted sulforaphane derivatives were simulated by Chem3D software, and the correlation of the NBO charge of carbonyl oxygen 5-O was found by fitting. The value was larger(R2 = 0.951, y=1 028.24 + 1 080.58 x), and the correlation coefficient of SGBP value of carbonyl oxygen 5-O fitted by information theory index was the largest(y=25.13 + 1.60 x, R2 = 0.980). The mass spectra of sulforaphane and its derivatives were simulated, and the characteristic peaks were identified and assigned. This paper can provide a solid theoretical support for the enzymatic synthesis of sulforaphane and its derivatives, as well as basic data for the identification of sulforaphane derivatives and their structure-activity relationships in materials engineering and pharmaceuticals.
