摘要
目的研究宫颈癌细胞泛素特异性蛋白酶22(USP22)表达水平及其与顺铂化疗敏感性的关系。方法采集宫颈癌细胞系Si Ha,以人永生化表皮细胞系Ha CaT为对照,通过实时荧光定量聚合酶链反应比较USP22 mRNA在两种细胞中的表达情况。将宫颈癌细胞系Si Ha分为对照组(转染USP22)和实验组(转染USP22-siRNA),采用CCK-8实验比较两组细胞增殖能力和细胞存活率,通过流式细胞仪检测细胞细胞周期和细胞凋亡率;采用裸鼠荷瘤实验,对照组给予磷酸盐缓冲液,实验组给予顺铂,检测宫颈癌肿瘤体积以评价顺铂化疗敏感性。结果相比人永生化表皮细胞系Ha CaT,宫颈癌细胞系Si Ha中USP22 mRNA相对表达量明显升高(P<0.01)。CCK-8实验结果显示,实验组宫颈癌Si Ha细胞增殖能力和存活率较对照组明显下降(P<0.01)。流式细胞仪检测结果显示,相比对照组,实验组宫颈癌Si Ha细胞G1期比率明显升高,S期细胞比率明显下降(P<0.01);两组G2期细胞比率的比较,差异无统计学意义(P>0.05)。实验组宫颈癌Si Ha细胞凋亡率较对照组明显升高(P<0.01)。裸鼠荷瘤实验结果发现,实验组肿瘤体积增长速度明显减缓(P<0.01)。结论USP22高表达于宫颈癌细胞,可有效诱导肿瘤细胞增殖,促使其对顺铂化疗的敏感性下降;沉默USP22在宫颈癌Si Ha细胞中的表达,具有明显抑制肿瘤细胞增殖、减少细胞存活、干扰细胞周期、促进细胞凋亡和缩小肿瘤体积的作用,同时可增强肿瘤细胞对顺铂化疗的敏感性。
Objective To study the expression of ubiquitin-specific protease 22(USP22)in cervical cancer cells and its relationship with the chemosensitivity of cisplatin.Methods Cervical cancer cell line SiHa was collected and compared with human immortalized epidermal cell line Ha CaT by real-time fluorescence quantitative polymerase chain reaction.Cervical cancer cell line Si Ha was divided into two groups:control group(transfected USP22)and experimental group(transfected USP22-siRNA).Cell proliferation and cell viability were compared by CCK-8 test.Cell cycle and apoptotic rate were detected by flow cytometry.Nude mice tumor-bearing experiment was conducted.Phosphate buffer was given to the control group,and cisplatin was given to the experimental group to detect the volume of cervical cancer to evaluate chemosensitivity of cisplatin.Results Compared with human immortalized epidermal cell line Ha CaT,the relative expression of USP22 in cervical cancer cell line SiHa was significantly increased(P<0.01).CCK-8 results showed that the proliferation ability and survival rate of Si Ha cells in the experimental group were significantly lower than those in the control group(P<0.01).The results of flow cytometry showed that compared with the control group,the proportion of SiHa cells in G1 phase was significantly increased and that of S phase was significantly decreased in the experimental group(P<0.01);there was no significant difference in the proportion of G2 phase cells between the two groups(P>0.05).The apoptotic rate of Si Ha cells in the experimental group was significantly higher than that in the control group(P<0.01).The growth rate of tumor volume in nude mice was significantly slowed down(P<0.01).Conclusion The high expression of USP22 in cervical cancer cells can effectively induce the proliferation of cancer cells and reduce their sensitivity to cisplatin chemotherapy.Silencing the expression of USP22 in SiHa cells of cervical cancer can significantly inhibit the proliferation of cancer cells,reduce cell survival,interfere with cell cycle,promote cell apoptosis and reduce the volume of cancer cells,and enhance the effect of cancer cells on cisplatin chemosensitivity.
