摘要
目的建立五味子标准汤剂中4种木脂素类成分的超高效液相色谱含量测定方法。方法应用UHPLC-DAD法,乙腈-水为流动相梯度洗脱,柱温30℃,流速0.45 mL·min^-1,检测波长220 nm,同时测定不同批次五味子标准汤剂中五味子醇甲、五味子醇乙、当归酰戈米辛H和五味子甲素4种木脂素类成分的含量。结果10批五味子标准汤剂中4种木脂素类成分的含量分别为:五味子醇甲0.1959%~0.3245%、五味子醇乙0.03574%~0.07154%、当归酰戈米辛H 0.02702%~0.05056%和五味子甲素0.01629%~0.04224%。结论建立了五味子标准汤剂中4种木脂素类成分的超高效液相色谱含量测定方法,该方法专属性强、准确度高、高效可靠,为五味子标准汤剂的质量评价提供了参考依据。
Objective To establish a simultaneous quantification method to determine four lignans in Schisandrae chinensis fructus standard decoction using ultra-high performance liquid chromatography(UHPLC)method.Methods The separation and detection procedure was performed on a Waters CORTECS T3 column(100×2.1 mm,2.7μm)at 30℃under a wavelength of 220 nm.The mobile phase was composed of water and acetonitrile using gradient elution at a flow rate of 0.45 mL·min^-1.Four lignans including schisandrin,schisandrol B,angeloylgomisin H and deoxyschisandrin were quantitative determined using the optimized UHPLC-DAD method.Results The contents of schisandrin,schisandrol B,angeloylgomisin H and deoxyschisandrin was 0.1959%-0.3245%,0.03574%-0.07154%,0.02702%-0.05056%,0.01629%-0.04224%,respectively.Conclusion The established UHPLC-DAD method is specifical,reliable and efficient and can be used to provide a reference standard for quality evaluation of Schisandrae Chinensis Fructus standard decoction.
