摘要
目的建立景天三七总黄酮的含量测定方法。方法通过方法适用性分析,选择适合景天三七的对照品及显色体系;对选出的AlCl3-KAc显色体系以星点设计-响应面法对其最佳显色剂配比进行优化,并进行方法学验证;对11批景天三七样品的总黄酮进行含量测定。结果适合景天三七的对照品为山柰酚,显色体系为AlCl3-KAc显色体系;星点设计-响应面法优化的AlCl3-KAc显色体系的最佳显色剂配比:AlCl3为0.5 mol·L-1、KAc为1.5 mol·L-1;11批景天三七样品总黄酮平均含量7.8 mg·g-1。结论建立的景天三七总黄酮含量测定方法实用性强,为黄酮的含量测定提供了新的参考方法。
Objective To established a method for determination of the content of total flavonoids in Sedum aizoon L..Methods Through applicability analysis,the reference substance and colour reaction system were selected;Optimize the color reaction ratio for the selected AlCl3-KAc colour reaction system by central composite design-response surface methodology,and validated the methodology.Finally,the content of total flavonoids in 11 batches of S.aizoon were determined.Results The reference substance suitable for S.aizoon is kaempferol,and the colour reaction system is AlCl3-KAc;The AlCl3-KAc colour reaction systems of optimum proportion of chromogenic reagent is that AlCl3 is 0.5 mol·L-1 and KAc is 1.5 mol·L-1.The average content of total flavonoids in 11 batches S.aizoon is 7.8 mg·g-1.Conclusion The established method for determining the total flavonoids of S.aizoon is practical,which provided a new method for the determination of total flavonoids.
