β-榄香烯介导HIF-1α对人食管ESD术后成纤维细胞的抑制作用 被引量:1

摘要 目的:研究β-榄香烯通过介导缺氧诱导因子-1α(HIF-1α)对人行食管内镜黏膜下剥离术(ESD)后成纤维细胞的抑制作用。方法:将ESD术后食管肉芽组织中提取、培养的成纤维细胞分为对照组(常规培养)、不同浓度β-榄香烯观察组(160μmol·L^(-1)、320μmol·L^(-1)、480μmol·L^(-1)、640μmol·L^(-1)的β-榄香烯处理的ESD成纤维细胞),给药0、24 h、48 h、72 h后,用细胞增殖毒性检测法检测成纤维细胞生长抑制率,用流式细胞术检测成纤维细胞凋亡率,用蛋白质印迹法检测对照组和480μmol·L^(-1)β-榄香烯观察组HIF-1α的表达水平。结果:β-榄香烯浓度增加可减缓成纤维细胞增殖,成纤维细胞的凋亡率随着β-榄香烯浓度的增加而增加,HIF-1α表达水平随着β-榄香烯浓度的增加而降低;用HIF-1α过表达质粒转染的成纤维细胞中的HIF-1α水平显著升高,通过480μmol·L^(-1)β-榄香烯处理可逆转升高。结论:β-榄香烯通过介导HIF-1α水平,减弱人食管成纤维细胞的增殖和加速成纤维细胞凋亡来抑制食管纤维化。 Objective To investigate the inhibitory effect ofβ-elemene on human esophageal fibroblasts after endoscopic submucosal dissection(ESD)by mediating hypoxia-inducible factor-1α(HIF-1α).Methods Fibroblasts extracted and cultured from esophageal granulation tissue after endoscopic submucosal dissection(ESD)were divided into a control group(conventional culture),and an observation group with different concentrations ofβ-elemene(160μmol·L^(-1),320μmol·L^(-1),480μmol·L^(-1),640μmol·L^(-1)ofβ-elemene treated ESD fibroblasts),and administered after 0,24 h,48 h and 72 h,the growth inhibition rate of fibroblasts was detected by cell proliferation toxicity assay,the apoptosis rate of fibroblasts was detected by flow cytometry,and the expression level of HIF-1αin the control group and 480μmol·L^(-1)ofβ-elemene observation group were detected by protein blotting.Results The proliferation of fibroblasts was inhibited by increasing the concentration ofβ-elemene,the apoptosis rate of fibroblasts was increased with the increasing concentration ofβ-elemene,and the expression of HIF-1αwas decreased.The level of HIF-1αin fibroblasts transfected with HIF-1αoverexpression plasmid was significantly increased,and the increase was reversed by 480μmol·L^(-1)β-elemene treatment.Conclusionβ-elemene inhibits esophageal fibrosis by mediating HIF-1αlevels,attenuating the proliferation of human esophageal fibroblasts and accelerating fibroblast apoptosis.
