摘要
目的:研究外周血中程序性死亡受体-1(PD-1)和干扰素γ(IFN-γ)在原发性肝癌(PHC)患者中的表达水平,探讨其在PHC发病中的作用。方法:选取自2018年8月至2020年8月于我院接受治疗的62例PHC患者为PHC组,同期46例慢性乙型肝炎(CHB)患者为CHB组,另选30例健康体检者为对照组。采用流式细胞术检测外周血单个核细胞(PBMC)中CD4^(+)T细胞和CD8^(+)T细胞中PD-1的表达,同时采用酶联免疫吸附测定(ELISA)法检测血清中IFN-γ水平,并分析PHC患者体内PD-1与IFN-γ的相关性。结果:与对照组和CHB组相比,PHC患者外周血PD-1表达及血清中IFN-γ水平均显著升高(P<0.01),且CHB组高于对照组,差异均具有统计学意义(均P<0.05)。且PHC患者体内PD-1的表达与IFN-γ水平呈正相关(r=0.526,P<0.05)。结论:PHC患者外周血中PD-1和IFN-γ的表达水平均升高,且PD-1的表达与IFN-γ水平呈正相关。
Objective:To investigate the expression levels of programmed cell death-1(PD-1)and interferonγ(IFN-γ)in peripheral blood in patients with primary hepatic carcinoma(PHC),and explore their role in the pathogenesis of PHC.Methods:62 patients with PHC who were diagnosed and treated in our hospital from August2018 to August 2020 were selected as PHC group,46 patients with chronic hepatitis B(CHB)during the same period as CHB group,another 30 healthy people were selected as control group.Flow cytometry was used to detect the expression of PD-1 in CD4^(+)T and CD8^(+)T cells in peripheral blood mononuclear cells(PBMC).The level of IFN-γin serum was detected by enzyme linked immunosorbent assay(ELISA),and the correlation between PD-1 and IFN-γwas analyzed in PHC group.Results:The expression of PD-1 and the level of IFN-γin PHC group were significantly increased than those in control group and CHB group,moreover,CHB group was higher than control group,the differences were all statistically significant(P<0.05).And the expression of PD-1 was positively correlated with IFN-γin PHC group(r=0.526,P<0.05).Conclusion:The expression level of PD-1 and the level of IFN-γin peripheral blood of PHC patients are increased and positively correlate.
