国产小型化流式细胞仪对HIV感染者CD4^+T淋巴细胞检测性能分析 被引量:2

摘要 目的 1)分析新一代小型化国产流式细胞仪平台与进口流式细胞仪平台对艾滋病病毒感染者CD4^+T淋巴细胞(简称CD4细胞)检测的准确性、相关性和一致性;2)比较同一样本由国产抗体试剂和进口抗体试剂处理后在FACS Calibur上CD4细胞的检测结果,分析抗体试剂对实验结果的影响。方法利用三种不同厂家的CD3/CD4/CD8/CD45四色抗体试剂平行处理100份感染者的血液样本,随后在Sparrow、FACS Calibur和FC500上进行CD4细胞检测,并对检测结果进行相关性分析、偏差分析及分层统计。结果 1)Sparrow与FACS Calibur和FC500CD4细胞绝对计数结果的相关系数(r)分别为0.962、0.963,相关性高,平均偏差分别为15个/μL、-33个/μL,并将检测结果按<200个/μL、200~499个/μL、> 499个/μL分层进行卡方检验,部分P值> 0.05,无显著性差异;2)国产抗体试剂和进口抗体试剂在FACS Calibur上CD4细胞检出结果的相关系数(r)为0.976,平均偏差为-16个/μL。结论 1)国产流式细胞仪对CD4细胞检测结果与进口仪器高度一致,相关性较高,所测定的绝对计数结果和设定的参考范围具有一定通用性;2)国产抗体试剂在进口流式细胞仪平台上也可适用,且与进口试剂处理结果高度一致。 Objective 1) To analyze the accuracy, correlation and consistency of CD4^+T lymphocyte detected by imported and domestic flow cytometry;2) To compare the CD4^+T lymphocyte count of a sample treated with domestic antibody reagent and imported antibody reagent. Methods Sparrow, FACS Calibur and FC500 were used to detect CD4^+ T lymphocyte counts of 100 samples which were parallelly treated with CD3/CD4/CD8/CD45 antibody reagents from HIV infected individuals, and to analyze the influence of antibody reagent and flow cytometry. Results 1) The correlation coefficient(r) of the CD4^+T lymphocytes between Sparrow vs FACS Calibur and Sparrow vs FC500 was 0.962, and 0.963 respectively, indicating high correlation, with the mean bias of 15 cells/μL, and-33 cells/μL respectively. The CD4^+ T lymphocyte count results were stratified into < 200 cells/μL, 200-499 cells/μL and > 499 cells/μL(P > 0.05), with no significant difference. 2) The correlation coefficient(r) of the CD4^+T lymphocytes, detected by domestic and imported antibody reagents in FACS Calibur, was 0.976, with a mean bias of-16 cells/μL. Conclusion 1) Data generated by the three types of flow cytometry are similar and highly correlated in CD4^+T lymphocyte detection. The absolute counting results and the reference range have a certain degree of universality. 2) Domestic antibody reagent can also be used by the imported flow cytometry, and the results are highly consistent with that of the imported antibody reagent.
