摘要
目的 构建拉莫三嗪血药浓度的遗传-反向genetic algorithm back propagation,GA-BP神经网络预测模型,初步为需要调整拉莫三嗪使用剂量或不能进行拉莫三嗪血药浓度监测的患儿提供浓度预测,可以根据个体化情况在临床中有效地制定给药方案。方法 收集自治区人民医院2015年10月-2021年4月就诊的规律服用拉莫三嗪15d以上癫痫患者病例总计201例。收集相关数据,构建GA-BP神经网络模型预测拉莫三嗪血药浓度。结果 32个样本的预测结果与实际浓度相比,平均预测误差(mean prediction error,MPE)为0.17%,平均绝对误差(mean absolute prediction error,MAE)为1.26%,32个预测误差(prediction error,PE)均小于10%,小于15%的概率为100%,血药浓度预测值与实测值之间的相关系数为0.9995,预测结果较理想。结论 反向传播神经网络可用于预测新疆癫痫患儿拉莫三嗪血药浓度,有待应用于临床,辅助个体化给药。
Objective To build a genetic algorithm-back propagation(GA-BP) neural network prediction model of lamotrigine serum concentration,and initially provide a concentration prediction model for children who need to adjust the dosage of lamotrigine or cannot monitor the serum concentration of lamotrigine.And dosing regimen can be effectively tailored in the clinic based on Individualized situation.Methods A total of 201 cases of epilepsy patients who regularly took lamotrigine for more than 15 days in the People’s Hospital of the Autonomous Region from October 2015 to April 2021 were collected.Relevant data were collected and GA-BP Neural Network Model were constructed to predict lamotrigine serum concentration.Results Compared with the actual concentration of the prediction results of 32 samples,the mean prediction error(MPE) was 0.17%,the mean absolute prediction error(MAE) was 1.26%,and32 prediction errors(PE) are all less than 10%,and the probability of less than 15% is 100%.The correlation coefficient between the predicted value of serum drug concentration and the measured value is 0.9995,and the predicted result is ideal.Conclusion GA-BP neural network can be used to predict the serum concentration of lamotrigine in children with epilepsy in Xinjiang,and it needs to be applied to clinical individualized administration.
