摘要
[目的]研究白鲜皮的肝保护活性成分.[方法]取干燥粉碎的白鲜皮用95%(体积分数)乙醇提取3次,用蒸馏水混悬乙醇提取浓缩液后依次用石油醚、乙酸乙酯和正丁醇萃取.利用硅胶柱色谱、YMC柱色谱、葡聚糖凝胶柱色谱法进行分离,通过核磁共振技术和质谱等波谱数据鉴定化合物结构.用叔丁基过氧化氢诱导HepG2细胞后用分离的化合物进行体外肝保护活性评价.[结果]从白鲜皮中分离纯化得到13个化合物,分别鉴定为β-谷甾醇(1)、白鲜碱(2)、前茵芋碱(3)、4,8-二甲氧基-3-(3-甲基-2-丁烯基)-2-喹啉酮(4)、柠檬苦素(5)、梣酮(6)、黄柏酮(7)、吴茱萸苦素(8)、fraxinellonone(9)、6-甲氧基-7-羟基-香豆素(10)、angustifolin(11)、11,12,13-三降愈创木-6烯-4β,10β-二醇(12)和胡萝卜苷(13).化合物7和8的肝保护EC50值分别为(53.23±0.34),(208.8±0.45)μmol/L.[结论]化合物11为首次从白鲜皮中分离得到的化合物;化合物7对t-BHP引起的HepG2细胞毒性的保护作用最好.
OBJECTIVE To study the hepatoprotective chemical constituents of Cortex Dictamni.METHODS Dried and pulverized Cortex Dictamni was extracted three times with 95%(volume fraction)EtOH.The EtOH extract was dissolved of H2O and then fractionated sequentially with petroleum ether,EtOAc and n-BuOH.The chemical constituents were isolated and purified by repeated column chromatography on silica gel,YMC and Sephadex LH-20 gel column chromatography,and their structures were elucidated by nuclear magnetic resonance technology and mass spectrum spectroscopic techniques.The hepatoprotective activity of the isolated compounds on t-BHP-induced cytotoxicity in HepG2 cells was evaluated.RESULTS Thirteen compounds were isolated from Cortex Dictamni which were elucidated asβ-sitosterol(1),dictamine(2),preskimmianine(3),glycolone(4),limonin(5),fraxinellone(6),obacunone(7),rutaevine(8),fraxinellonone(9),scopoletin(10),angustifolin(11),11,12,13-trinorguai-6 ene-4β,10β-diol(12),and daucosterol(13).Compounds 7 and 8 showed significant hepatoprotective effects with the EC50values of(53.23±0.34)μmol/L and(208.8±0.45)μmol/L,respectively.CONCLUSION Compound 11 was isolated from Cortex Dictamni for the first time.Compound 7 had the best protective effect on t-BHP-induced HepG2 cytotoxicity.
