摘要
[目的]探讨紫檀芪对视网膜色素上皮细胞损伤的保护作用机制.[方法]建立蓝光诱导负载N-亚视黄基-N-视黄基乙醇胺(A2E)的人视网膜色素上皮细胞(ARPE-19)光损伤模型.利用MTT比色法检测紫檀芪(0.625、1.250、2.500、5.000μmol/L)对ARPE-19细胞的毒性;通过倒置显微镜观察紫檀芪对ARPE-19细胞损伤的影响;采用RT-qPCR法检测ARPE-19细胞凋亡相关基因CASP3和PARP1的表达水平.[结果]紫檀芪在5.000μmol/L浓度下对细胞无明显毒性,对蓝光负载A2E导致的ARPE-19细胞损伤及其CASP3和PARP1基因的表达具有显著的抑制作用(P<0.05,P<0.01).[结论]紫檀芪可通过抑制CASP3和PARP1基因的表达而发挥对ARPE-19细胞损伤的保护作用.
OBJECTIVE To explore the protective mechanism of pterostilbene on retinal pigment epithelial cell injure.METHODS Blue light-induced N-retinylidene-N-retinyletha-nolamine(A2 E)-loaded adult retinal pigment epithelial cells(ARPE-19)light damage model was established.MTT assay was used to detect the toxicity of pterostilbene(0.625,1.250,2.500,5.000μmol/L)on ARPE-19 cells.Inverted microscope was used to observe the effect of pterostilbene on ARPE-19 cells damage.RT-qPCR was used to detect the expression levels of apoptosis-related genes CASP3 and PARP1 in ARPE-19 cells.RESULTS Pterostilbene has no significant toxicity to cells under the concentration of 5.000μmol/L or less,and it significantly inhibited the damage of blue light-induced A2 E loaded ARPE-19 cells and significantly inhibited the expressions of CASP3 and PARP1 in blue light-induced A2 E loaded ARPE-19 cells(P<0.05,P<0.01).CONCLUSION Pterostilbene can protect ARPE-19 cells from damage by inhibition the expressions of CASP3 and PARP1.
