摘要
目的:研究香砂纯露和水提物含药血清对HSC-T6细胞的增殖和凋亡的作用。方法:取18只健康家兔随机分为3组,对照组,香砂纯露组(14.3 g/kg,剂量按生药量计),香砂水提物(50 g/kg),连续灌胃6天后,动脉采血,分离血清。结晶紫法用于检测香砂纯露和水提物含药血清对HSC-T6细胞的细胞毒性,作用48 h后,肉眼观察和拍照分析,并检测吸光度A值,然后,计算细胞增殖抑制率。Hoechst 33258染色用于分析香砂纯露和水提物含药血清对HSC-T6细胞的细胞凋亡。结果:与对照组比较,经香砂纯露和水提物含药血清处理48 h后,HSC-T6细胞数目明显减少,吸光度A值明显降低(P<0.01),细胞生长抑制率明显升高(P<0.01),细胞生长抑制率分别为62.29%和42.65%,细胞凋亡数目显著增加(P<0.01),细胞凋亡率显著升高(P<0.01),细胞凋亡率分别为41.25%和21.28%。结论:香砂纯露和水提物含药血清对HSC-T6有良好的生长抑制和促凋亡作用。
Aim:To study the effects of Water extract or Hydrosol from Aplinia zerubet Fruits containing serum on proliferation and apoptosis of HSC-T6 cells.Methods:Eighteen healthy rabbits were randomly divided into three groups,control group,Hydrosol from Aplinia zerubet Fruits group(14.3 g/kg),Water extract from Aplinia zerubet Fruits(50 g/kg),after six consecutive days administion,arterial blood collected,and the drug-containing serum separated.Crystal Voilet Staining was used to measure the cytotoxicity of 20%Water extract or Hydrosol from Aplinia zerubet Fruits containing serum,incubated for 48 h,visual examination and photo taking were used for analyzing,and A value determined,subsequently,their growth inhibition ratio calculated.Hoechst 33258 staining was used to measure apoptosis of HSC-T6 cells,visual examination and photo taking were used for analyzing,their apoptotic number counted,and the apoptotic ratio calculated.Results:Compared with control group,after treated with 20%Water extract or Hydrosol from Aplinia zerubet Fruits containing serum for 48 h,the numbers of HSC-T6 cells was obviously decreased,the A value of HSC-T6 cells was significantly decreased(P<0.01),and the growth inhibition rates was significantly increased(P<0.01),the growth inhibition rates of HSC-T6 cells treated with 20%Water extract and Hydrosol from Aplinia zerubet Fruits containing serum were 62.29%and 42.65%,the apoptotic numbers of HSC-T6 cells was significantly increased(P<0.01),and the apoptotic ratio was significantly increased(P<0.01),the apoptotic ratio of HSC-T6 cells treated with 20%Water extract and Hydrosol from Aplinia zerubet Fruits containing serum were 41.25%and 21.28%.Conclusions:Water extract or Hydrosol from Aplinia zerubet Fruits containing serum has good growth inhibition effect and promoting apoptosis effect on HSC-T6 cells.
