基于网络药理学的夏枯草治疗Graves病机制探讨 被引量:9

摘要 目的:运用网络药理学方法筛选并预测夏枯草治疗Graves病(GD)的关键成分、核心靶点并探讨其可能的作用机制。方法:通过多个数据库查询筛选夏枯草的活性成分及其治疗GD的人源靶标,通过Cytoscape 7.1软件分别构建成分-靶标网络、夏枯草治疗Graves病蛋白互作网络,利用网络拓扑分析筛选夏枯草治疗GD核心靶点,采用ClueGO插件进行KEGG基因富集分析,得到夏枯草治疗GD的可能作用通路。结果:共筛选出14个夏枯草活性成分,98个夏枯草与Graves病共有靶点,网络拓扑分析最终筛选出夏枯草治疗GD的12个核心靶点(TNF、IL6、TP53、IL10、VEGFA、JUN、STAT3、CCL2、IL1β、IL4、CXCL8、EGFR),β-谷甾醇、山奈酚、木樨草素、槲皮素、芦丁、熊果酸、迷迭香酸7种关键化合物,通过对12个核心靶点的富集分析筛选出IL-17信号通路、Toll样受体信号通路、TNF通路、HIF-1信号通路等6条夏枯草治疗GD的信号通路。结论:夏枯草可通过作用于VEGF靶点抑制GD患者的甲状腺肿大,预测IL-17信号通路、Toll样受体信号通路、TNF信号通路为夏枯草治疗GD的潜在通路。 Objective:To screen and predict the key components and core targets of Prunella Vulgaris treating Graves disease(GD)using network pharmacology and to explore its possible mechanism.Methods:The active components of Prunella Vulgaris and human target of GD were screened through multiple database queries.The component-target network and the protein interaction network of Prunella Vulgaris treating Graves disease were constructed by Cytoscape 7.1 software,and the core targets were screen out by the network topology analysis.Then ClueGO plug-in was used for KEGG gene enrichment analysis to obtain a possible pathway for Prunella vulgaris treating GD.Results:A total of 14 active components of Prunella Vulgaris,98 common targets of Prunella Vulgaris and Graves disease were screened out,and the network topology analysis finally screened out 12 core targets(TNF,IL6,TP53,IL10,VEGFA,JUN,STAT3,CCL2,IL1β,IL4,CXCL8,EGFR),β-sitosterol,kaempferol,luteolin,quercetin,rutin,ursolic acid,rosmarinic acid.Through the Enrichment analysis of 12 core targets six signal pathways for prunella vulgaris treatment were screened,which are IL-17 signaling pathway,Toll-like receptor signaling pathway,TNF pathway,and HIF-1 signaling pathway.Conclusion:Prunella Vulgaris can inhibit the enlargement of thyroid goiter in patients with GD by acting on VEGF targets.Furthermore,IL-17 signaling pathway,Toll-like receptor signaling pathway and TNF signaling pathway are possible pathways for prunella treating GD.
