从网络药理学角度探究骨碎补治疗骨折类疾病的作用机制 被引量:5

Exploration of the Mechanism of Rhizoma Drynariae in the Treatment of Fracture Diseases from the Perspective of Network Pharmacology

摘要 目的:基于网络药理学分析骨碎补中有效成分治疗骨折类疾病的作用机制。方法:(1)从TCMSP数据库中搜索中药骨碎补的有效活性成分,以OB≥30%和DL≥0.18为筛选条件,得到骨碎补中活性成分及其对应靶基因;(2)分别在GeneCards数据库和OMIM数据库中筛选骨折对应靶基因;(3)取骨碎补靶基因和骨折靶基因的交集,得到骨碎补-骨折共同靶基因并构建中药调控网络;(4)利用软件Cytoscape构建"骨碎补-活性成分-靶基因-骨折"调控网络;(5)利用string软件构建PPI网络,并找出核心基因;(6)对骨碎补-骨折共同靶基因进行GO功能富集分析和KEGG通路富集分析。结果:骨碎补作用于骨折的有效活性成分有14个,骨碎补-骨折共同靶基因55个,共同靶基因组成73种功能富集以及101条网络通路。结论:研究结果显示骨碎补在治疗骨折类疾病时,通过多成分、多靶基因、多通路的共同作用显效,为骨碎补促进骨折愈合的作用机制研究提供了参考依据。 Objective:To analyze the action mechanism of the active ingredients in rhizoma drynariae in the treatment of fracture diseases based on network pharmacology.Methods:(1)Search the effective active ingredients of Chinese medicine rhizoma drynariae from TCMSP database,use OB≥30%and DL≥0.18 as the screening conditions to obtain the active ingredients and corresponding target genes of rhizoma drynariae;(2)Screen the target genes for fractures in the GeneCards database and the OMIM database;(3)Take the intersection of rhizoma drynariae target gene and fracture target gene to obtain the rhizoma drynariae-fracture common target gene and construct a Chinese medicine regulatory network;(4)Use the software Cytoscape to build a"Rhizoma drynariae-Active Ingredient-Target Gene-Fracture"regulatory network;(5)Use string software to build a PPI network and find out the core genes;(6)GO function enrichment analysis and KEGG pathway enrichment analysis were obtained from the common target genes of rhizoma drynariae and fracture.Results:Rhizoma drynariae has 14 effective active ingredients for fractures.There are 55 rhizoma drynariaefracture common target genes,and constitute of 73 functional enrichment,and 101 network pathways.Conclusion:The research results in this article show that rhizoma drynariae is effective in the treatment of fracture diseases through the combined effects of multiple components,multiple target genes,and multiple pathways,providing further research basis for the mechanism of rhizoma drynariae to promote fracture healing.
