摘要
目的:制备盐沙苑子标准汤剂并建立质量标准。方法:制备15批盐沙苑子饮片标准汤剂,以沙苑子苷为检测指标,计算指标成分含量和转移率,建立超高效液相指纹图谱,采用《中药色谱指纹图谱相似度评价系统(2012版)》软件进行相似度评价,采用UPLC-Q-TOF-MS/MS对共有峰进行结构鉴定。结果:盐沙苑子饮片标准汤剂中沙苑子苷转移率范围为42.93%~81.22%,平均转移率为60.10%,15批盐沙苑子饮片标准汤剂指纹图谱与对照指纹图谱相似度均>0.9。指纹图谱共有峰13个,确认13个,分别为芒柄花黄素、杨梅素、萹蓄苷、杨梅素-3-O-β-D-葡萄糖苷、毛蕊异黄酮-7-O-β-D-葡萄糖苷、山柰酚3-O-β-D-葡萄糖苷、橘皮素、西伯利亚落叶松黄酮、沙苑子苷、愈创木基甘油-β-阿魏酸醚、毛蕊异黄酮、大麻苷、4 H-1-苯并吡喃-4-酮、3-((6-O-乙酰基-β-D-吡喃葡萄糖基)氧基)-5-羟基-2-(4-羟基苯基)-7-甲氧基。结论:该研究建立的盐沙苑子标准汤剂制备工艺稳定,样品来源具有代表性,建立的指纹图谱方法精密度、稳定性及重复性良好,可为盐沙苑子配方颗粒的质量控制提供参考。
Objective:To prepare standard decoction of salt Complanatus,establish quality standards.Methods:Prepare 15 batches standard decoction of salt Complanatus pieces,With Complanatoside as the detection index,the extraction rate,content and transfer rate of index components were calculated and the UPLC characteristic chromatograms was established.Similarity Evaluation System for Chromatographic Fingerprint of TCM(2012)was used to analyze and compare the characteristic chromatograms.The chemical structures of common peaks were identified with ultra-performance liquid chromatography with quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)in the standard decoction of salt Complanatus.Results:The result revealed that the extraction ratio was from 42.93%to 81.22%,with an average value of 60.10%,fingerprint and the similarity between the fingerprint of standard decoction of salt Complanatus and the reference fingerprint was over 0.9.There are 13 peaks in the fingerprint,and 13 are confirmed,respectively,Formononetin,Myricetin,Guajavarin,myricetin-3-O-β-D-glucopyranoside,calycosin-7-O-β-D-glucoside,kaempferol-3-O-β-D-glucoside,Tangeretin,Laricitrin,complanatuside,Guaiacylglycerol-β-ferulic acid ether,Calycosin,cannabiscitrin,4 H-1-Benzopyran-4-one,3-((6-O-acetyl-β-D-glucopyranosyl)oxy)-5-hydroxy-2-(4-hydroxyphenyl)-7-methoxy.Conclusion:The preparation method of salt Complanatus standard decoction established in this study is stable,the sample source is representative,and the established fingerprint method has good precision,stability and repeatability,which can provide reference for the quality control of salt Complanatus formula particles.
