摘要
胃得安是一种由25种中草药配制而成的传统中药,主要用于治疗胃痛和胃酸分泌过多。为探究胃得安是否对胃溃疡具有保护作用,作者选用ICR小鼠,连续2 w灌胃不同剂量的胃得安,同时以三九胃泰作为阳性对照,再用乙醇建立急性胃溃疡模型。通过胃组织解剖评分来评估胃黏膜的外观和形态损伤;苏木精-伊红染色观察胃组织中的炎性细胞浸润;检测丙二醛(MDA),超氧化物歧化酶(SOD)和髓过氧化物酶(MPO)等氧化应激标志物;分别通过实时定量PCR(RT-qPCR)和酶联免疫吸附试验(ELISA)检测炎症相关的细胞因子,包括IL-6、TNF-α、IL-10和血清C反应蛋白(CRP)。结果显示,胃得安给药组小鼠胃溃疡面积、胃溃疡指数及胃组织中免疫细胞的浸润均显著减少。同时,与阴性对照组相比,不同剂量胃得安给药后小鼠胃组织的MPO和MDA水平均有一定程度的降低,SOD水平升高,并呈剂量依赖性。另外给药后小鼠胃组织的炎性相关细胞因子如IL-10、IL-6和TNF-α水平均有下降,血清CRP减少。初步证明了胃得安对乙醇诱导的胃溃疡具有保护作用,其机理可能与胃得安具有抗氧化和抗炎作用相关。
Wei Dean,a prescription of traditional Chinese medicine containing 25 herbal ingredients,has been used to treat stomac-hache and excessive secretion of stomach acid.To investigate its potential protective effect against gastric ulcer,the authors estab-lished ethanol-induced acute gastric ulcerin ICR mice.Before the establishment of acute gastric ulcer,mice were orally administrated with Wei Dean at different dose every day for 2 weeks.The gross appearance and morphological damage of gastric mucosa were evaluated by dissecting gastric tissue,and inflammatory cell infiltration was observed by hematoxylin-eosin(HE)staining of the gas-tric tissue.The authors also measured oxidative stress markers,such as malondialdehyde(MDA),superoxide dismutase(SOD),and myeloperoxidase(MPO),as well as inflammation-related cytokines,including IL-6,TNF-α,IL-10 and serum C-reactive protein(CRP)by real-time quantitative PCR(RT-qPCR)and enzyme-linked immunosorbent assay(ELISA),respectively.The results showed that Wei Dean administration reduced the gastric ulcer area and gastric ulcer index,and reduced the infiltration of immune cells in gastric tissue.In addition,the levels of MPO and MDA were significantly lower in Wei Dean-pretreated mice compared to the negative control,while the level of SOD was higher.Furthermore,the levels of inflammatory cytokines,including IL-10,IL-6,and TNF-α,were reduced,as well as the serum CRP.In conclusion,the current study illustrated that Wei Dean has significant preventive effects against ethanol-induced gastric ulcers,and its mechanisms of action involve antioxidant and anti-inflammatory properties.
