摘要
为研究杂交籼稻骨干亲本Wx和ALK基因的等位变异与水稻蒸煮食味品质的关联性及其理想组合方式,对34份杂交籼稻骨干亲本的稻米直链淀粉含量(AC)、胶稠度(GC)、碱消值(ASV)与其Wx和ALK基因等位变异的关联性进行分析.结果显示:34份材料的Wx基因序列包括41个多态性位点,被划分为6种单倍型,共有31个位点与AC和GC呈极显著关联,其中Ex1-47位点的关联系数最大.Wx1单倍型在Int1-1位点与Wx^(a)相同,与中高AC和硬GC极显著关联;Wx2单倍型在Int1-1和Ex1-47位点与Wx^(hp)相同,与中高AC和中等GC极显著关联;Wx3、Wx4、Wx5和Wx6单倍型在Int1-1位点与Wx^(b)相同,与较低AC和软GC极显著关联.34份材料的ALK基因包括49个多态性位点,被划分为5种单倍型,共有24个位点与ASV呈显著或极显著关联,其中Ex8-864/865位点的关联系数最大.ALK1、ALK2、ALK3和ALK4单倍型在Ex8-733和Ex8-864/865位点与ALK^(G)一致,其中ALK1、ALK3和ALK4与低ASV极显著关联,ALK2与中高ASV极显著关联.ALK 5单倍型在Ex8-733和Ex8-864/865位点与ALK^(T)一致,与高ASV极显著关联.34份材料共分为10个Wx和ALK基因单倍型组合,不同单倍型组合间的AC、GC和ASV均存在显著或极显著差异,其中Wx3-ALK5和Wx5-ALK5单倍型组合表现为较低AC、软GC和高ASV.本研究表明Wx3-ALK5和Wx5-ALK5单倍型组合是优质稻米蒸煮食味品质的理想组合方式.
To identify the best allelic combination of the Wx and ALK genes from elite parents of hybrid rice,an association analysis between allelic variations in Wx and ALK genes and amylose content(AC),gel consistency(GC),and alkali spreading value(ASV)was performed on 34 elite parents of hybrid rice.The results showed that the Wx gene sequences of the 34 lines included 41 polymorphic loci and could be classified into six haplotypes.Of these,31 polymorphic loci were significantly correlated with AC and GC,and the R^(2)value between the Ex1-47 variation site was the highest.The Wx1 haplotype was the same as Wx^(a)at the Int1-1 variation site and was significantly associated with medium-high AC and hard GC.The Wx2 haplotype was the same as Wx^(hp)in the Int1-1 and Ex1-47 variation sites and was significantly correlated with medium-high AC and intermediate GC.The Wx3,Wx4,Wx5,and Wx6 haplotypes were the same as Wx^(b)at the Int1-1 variation site and were significantly correlated with low AC and soft GC.The ALK sequences of the 34 lines included 49 polymorphic sites and were classified into five haplotypes.Among these 49 sites,24 were significantly or extremely significantly correlated with ASV,and the R^(2)value of the Ex8-864/865 variation site was the highest.The ALK1,ALK2,ALK3,and ALK4 haplotypes were consistent with ALK^(G)at the Ex8-733 and Ex8-864/865 variation sites.The ALK1,ALK3,and ALK4 haplotypes were significantly correlated with low ASV,whereas ALK2 was significantly correlated with medium-high ASV.The Ex8-733 and Ex8-864/865 variation sites in the ALK5 haplotype had the same mutation sites as those of ALK^(T),which was significantly correlated with high ASV.There were ten haplotype combinations in the Wx and ALK genes of 34 parents.The AC,GC,and ASV values of the different haplotype combinations were significantly or extremely significantly different.The Wx3-ALK5 and Wx5-ALK5 haplotype combinations were associated with low AC,soft GC,and high ASV.Therefore,the Wx3-ALK5 and Wx5-ALK5 haplotype combinations were ideal combinations for good cooking and eating quality for parents of hybrid rice.
