火针联合回阳玉龙膏对局限性硬皮病鼠模型TGF-β1/Smad信号通路的影响 被引量:3

Effects of Fire Needle Combined with Huiyang Yulong Ointment on TGF-β1/Smad Signaling Pathway in a Rat Model of Localized Scleroderma

摘要 目的 观察火针联合回阳玉龙膏对局限性硬皮病鼠模型TGF-β1/Smad信号通路的影响。方法 将36只SPF级BALB/c小鼠随机分为A组、B组、C组、D组、E组和F组,共6组,每组6只。除A组外,其余5组均建立小鼠硬皮病模型,造模28 d。造模结束后,A组及B组不给予任何干预,其余4组分别给予肝素钠软膏、回阳玉龙膏外用、火针针刺及火针联合回阳玉龙膏4种方法治疗28 d。肉眼观察各组小鼠皮损,镜下观察组织病理学变化,Masson染色法评估各组小鼠皮肤纤维化指数,免疫组织化学染色法检测各组小鼠皮肤中TGF-β1、LIGHT、Smad3、Smad7的表达含量,偏正光皮肤镜检测各组小鼠皮肤组织表面血管毛发的变化情况,甚高频皮肤超声检测各组小鼠皮肤组织厚度的变化情况。结果 博莱霉素诱导的硬皮病小鼠模型纤维化指数明显升高,皮肤中TGF-β1、LIGHT、Smad3表达增加,Smad7表达减少。治疗28 d后,回阳玉龙膏外用、火针针刺及二者联合均可不同程度的减轻博莱霉素致硬皮病小鼠的皮肤真皮层增厚及纤维化指数,与B组比较,D组、E组、F组的TGF-β1、LIGHT、Smad3的含量均有所下降,Smad7含量升高,真皮层厚度明显变薄,尤以F组效果明显(P<0.05)。结论 火针针刺、回阳玉龙膏外用以及火针联合回阳玉龙膏三种方法可能通过抑制TGF-β1/Smad通路降低TGF-β1、LIGHT、Smad3、增加Smad7的表达水平来缓解硬皮病组织纤维化的过程。回阳玉龙膏外用联合火针针刺治疗局限性硬皮病的疗效优于单用火针或回阳玉龙膏,协同作用增强疗效,具有起效更快、疗程更短、疗效更显著的特点。 Objective To observe the effect of fire needle combined with Huiyang Yulong ointment on TGF-β1/Smad signaling pathway in rat model of localized scleroderma.Methods Thirty-six SPF BALB/c mice were randomly divided into group A, group B, group C, group D, group E and group F. Except group A, the other 5 groups established mouse scleroderma models for 28 days. Group A and group B did not give any intervention, the other 4 groups were given Heparin sodium ointment, Huiyang Yulong ointment for external use, fire acupuncture and fire acupuncture combination with Huiyang Yulong ointment for 28 days respectively. The skin lesions of each group were observed by naked eyes and the expression of TGF-β1, LIGHT, Smad3, Smad7 were detected by immunohistochemistry and skin fibrosis index was evaluated by Masson staining, the changes of blood vessel and hair on the skin tissue were detected by polarized light dermatoscope and the changes of skin tissue thickness were detected by very high frequency skin ultrasound.Results The fibrosis index of bleomycin-induced scleroderma mouse model was significantly increased, the expression of TGF-β1,LIGHT and Smad3 was increased, and the expression of Smad7 was decreased. After external application of Huiyang Yulong ointment, fire-needle acupuncture and combined treatment for 28 days, Masson staining showed that the fibrotic index of mice in each group decreased obviously, and the dermis layer became thinner obviously. Compared with group B,the expressions of TGF-β1, LIGHT and Smad3 were decreased, and the expression of Smad7 was increased in group D,E,F. Especially in group F, the effect was obvious(P<0.05).Conclusion The combination of external application, fire acupuncture and the combination of the two may alleviate the process of scleroderma fibrosis by inhibiting the TGF-β1/Smad pathway, decreasing the expression of TGF-β1, LIGHT, Smad3 and increasing the expression of Smad7. The curative effect of external application of Huiyang Yulong ointment combined with fire needle acupuncture is better than that of single application of fire-needle or Huiyang Yulong ointment.
