摘要
目的构建中重症EV71型手足口病的BABL/c乳鼠模型。方法1日龄BALB/c乳鼠连续3 d腹腔注射EV71病毒液,监测小鼠体质量、生存率和临床疾病评分;采用HE染色检测脑、肺和骨骼肌组织病理变化;光镜下观察感染乳鼠脑、肺和骨骼肌组织匀浆作用RD细胞的细胞病变反应;采用蛋白质免疫印迹和免疫荧光法检测VP1和3D蛋白水平。结果与正常对照组相比,EV71感染乳鼠体质量从第3天开始下降,2组比较差异有统计学意义(t=8.4210,P<0.05);生存率随感染时间延长逐渐下降;疾病评分显著增加;脑、肺及骨骼肌组织中VP1和3D蛋白表达与正常对照组比较显著升高(脑组织:t=6.3557、3.4232,肺组织:t=14.8648、4.4690,骨骼肌:t=7.9085、4.9237,均P<0.01)。结论成功构建了中重症EV71型手足口病BALB/c乳鼠模型。
Objective To construct BABL/c suckling mouse models of moderate and severe EV71 hand-foot-mouth disease.Methods The weight,survival rate and clinical disease score of 1-day-old BALB/c suckling mice were monitored after intraperitoneal injection of EV71 for 3 days.HE staining was used to observe the pathological changes of brain,lung and skeletal muscle.The RD cells were infected with homogenate of brain,lung and skeletal muscle tissues to observe the cytopathic effect(CPE).Western blotting and immunofluorescence analysis were used to detect the expression of VP1 and 3D proteins.Results Compared with NC group,the weight of EV71 infected suckling mice decreased from the 3rd day(t=8.4210,P<0.05);the survival rate of EV71 infected suckling mice decreased gradually with the extension of infection time,but the disease score increased significantly.In addition,compared with the normal group,the expression of VP1 and 3D proteins increased significantly than that in brain,lung and skeletal muscle respectively(t=6.3557,t=3.4232,t=14.8648,t=4.4690,t=7.9085,t=4.9237,all P<0.01).Conclusion The BALB/c suckling mouse models of moderate and severe EV71 hand-foot-mouth disease was successfully constructed.
