摘要
通过PD培养基对一株镰刀属(Fusarium sp.)的虎杖(Polygonum cuspidatum)内生真菌其进行发酵培养,采用硅胶、凝胶Sephadex LH-20、ODS-C18柱层析分离其发酵产物乙酸乙酯萃取物的化学成分进行分离,利用质谱和核磁共振等手段进行结构分析,鉴定出5个化合物,分别是肉桂酸(cinnamic acid),麦角甾醇(ergosterol),镰刀菌酸(fusaric acid),9,10-脱氢镰刀菌酸(9,10-dehydrofusaric acid),麦角甾-4,6,8(14),22-四烯-3-酮(ergosta-4,6,8(14),22-tetraen-3-one)。采用海虾致死实验对其提取物的细胞毒活性进行测试,石油醚、乙酸乙酯、正丁醇和水的提取相对丰年虾均有致死作用,致死率分别为66.47%、78.78%、57.62%和40.53%。
To ferment and culture endophytic fungus of Polygonum cuspidatum of Fusarium sp.on PD medium,separate chemical components of fermentation products of ethyl acetate extract with silica gel,gel Sephadex LH-20,ODS-C18 column chromatography,analyze structure with mass spectrometry and nuclear magnetic resonance.Five compounds were identified,namely cinnamic acid,ergosterol,and fusaric acid,9,10-dehydrofusaric acid(9,10-dehydrofusaric acid),ergot-4,6,8(14),22-tetraen-3-one(ergosta-4,6,8(14),22-tetraen-3-one).Test cytotoxic activity of extracts with shrimp lethal experiment.Extraction of petroleum ether,ethyl acetate,n-butanol and water all had lethal effects on brine shrimp,with lethal rates of 66.47%,78.78%,57.62%and 40.53%.
