摘要
目的探究基质细胞衍生因子-1(SDF-1)在急性肝衰竭(ALF)大鼠中的表达及意义。方法将20只雄性SD大鼠随机分为空白组和模型组,模型组采用TAA制备ALF模型,记录两组大鼠48 h生存率。48 h后腹主动脉采血,采集肝脏组织。检测两组大鼠ALT、AST、TBIL变化,HE染色观察肝组织病理变化,ELISA检测血清中SDF-1表达,免疫组化法检测肝组织中SDF-1表达。结果ALF大鼠血清SDF-1的浓度较正常组大鼠低(P<0.05),肝组织SDF-1表达较正常组明显增高(P<0.05),差异具有统计学意义。正常肝组织仅在汇管区及胆管上皮细胞弱阳性表达SDF-1。模型组大鼠肝脏显著表达SDF-1,主要位于胆管上皮及卵圆细胞,成熟肝细胞未见明显表达。结论ALF大鼠肝组织高表达SDF-1,血清低表达SDF-1,形成了有助于骨髓间充质干细胞(BMSCs)归巢的浓度差,可能是BMSCs归巢到肝脏发挥其修复作用的重要因素,同时,该浓度差可能是肝脏自然代偿性修复的重要机制之一。
Objective To explore the expression and significance of stromal cell-derived factor-1(SDF-1)in rats with acute liver failure(ALF).Methods Twenty male SD rats were randomly divided into two groups:the blank group and the model group.The ALF model was made by TAA in the model group,and the 48 h survival rate of the two groups was recorded.48 hours later,blood was collected from abdominal aorta and liver tissues were collected.The changes of alt,AST and TBIL,the pathological changes of liver tissue,the expression of SDF-1 in serum and the expression of SDF-1 in liver tissue were detected by HE staining,ELISA and immunohistochemistry.Results The serum SDF-1 concentration of ALF rats was lower than that of normal rats(P<0.05),and the expression of SDF-1 in liver tissue was significantly higher than that of normal rats(P<0.05).SDF-1 was only weakly expressed in the portal area and bile duct epithelial cells in normal liver tissue.In the model group,SDF-1 was significantly expressed in the liver,mainly in bile duct epithelium and oval cells,but not in mature hepatocytes.Conclusion High expression of SDF-1 in liver tissue and low expression of SDF-1 in serum of ALF rats may contribute to the difference of concentration of bone marrow mesenchymal stem cells(BMSCs)homing,which may be an important factor for BMSCs homing to the liver to play its repair role.At the same time,the difference of concentration may be one of the important mechanisms of liver natural compensatory repair.
