摘要
目的探讨2型糖尿病人群OX40的表达水平与MAIT细胞比例的相关性及凋亡作用,分析其对糖尿病的调控作用。方法收集51例2型糖尿病患者(T2D)和44例正常对照者的外周血,分离外周血单个核细胞,并采用流式细胞术检测MAIT细胞的比例和OX40分子的表达,比较OX40阴性和阳性MAIT细胞中CD45RO、CD69、Fas的表达,分析OX40表达与MAIT细胞比例的相关性及对其的凋亡作用。结果 T2D组MAIT细胞表达明显低于健康对照组;T2D患者循环的OX40+MAIT细胞比例增高;OX40+MAIT细胞表面CD45RO、CD69、Fas的比例明显高于OX40-MAIT细胞;相关性分析显示OX40表达与MAIT细胞呈负相关;OX40L可显著增强MAIT细胞凋亡。结论 OX40在T2D患者外周血循环的MAIT细胞上高表达,并与记忆和活化表型相关,且与MAIT细胞比例呈负相关,并促进MAIT细胞凋亡。OX40 MAIT细胞可能为2型糖尿病免疫活化的关键调控分子,有望成为2型糖尿病的潜在治疗靶点。
Objective To explore the expression of OX40 in mucosal associated invariant T cells (MAIT) of patient with type 2 diabete (T2D),also analyze correlation and apoptosis between OX40 and MAIT cells.Methods 51 cases of patients with type 2 diabetes and 44 cases of normal control subjects were collected.The peripheral blood mononuclear cell (PBMC) were isolated by ficoll method,and was detected by flow cytometry.To analysis the correlation between expression of OX40 and MAIT cells proportion and its role in apoptosis.The expression of CD45RO,CD69 and Fas in the OX40 negative and positive MAIT cells was compared.Results OX40 expression in MAIT cells in T2D group was obviously higher than that in healthy control group.The frequency of MAIT cell in T2D patients was reduced compared with healthy controls.OX40+MAIT cells expressed higher levels of CD45RO,CD69 and Fas than those in OX40-MAIT cells.OX40 expression was negative correlative with MAIT cells proportion in T2D patients;OX40L can significantly enhance MAIT cells apoptosis.Conclusion OX40 expression in MAIT cells was upregulated in T2D patients,and was associated with activation and effector phenotype,and promote MAIT cells apoptosis.OX40 may play an important role in immune regulation of MAIT cells in T2D development,which may be used as potential therapeutic target for type 2 diabetes.
