摘要
目的:用HPLC-ELSD建立蒙成药"三子散"中川楝素含量测定方法。方法:参照《中国药典》2015年版川楝子中川楝素含量测定的提取方法,采用HPLC-ELSD仪器,建立川楝素含量测定方法,测定三子散中川楝素的含量。建立数学模型,攻克蒸发光检测器计算双峰物质含量测定的难题,摸索色谱条件,并开展方法学考察。结果:HPLC-ELSD色谱条件确定为:C18柱,以乙腈-水(30∶70)为流动相,流速:1.0 mL/min,柱温:45℃,检测器漂移管温度:100.0℃,气体流速:2.8 L/min。方法学考察表明该方法有较好的精密度、准确度、重复性,在本研究条件下,川楝素在3.42~10.70μg内线性关系良好(r^(2)=0.995),平均回收率为97.2%,RSD=1.3%(n=6)。结论:本研究建立了HPLC-ELSD测定蒙成药"三子散"中川楝素含量的方法,该方法灵敏、快速、准确,适用于三子散中川楝素的含量测定。
Objective:To establish a method for the determination of toosendanin in Mongolian patent medicine Sanzisan by HPLC-ELSD.Methods:Referring to the extraction method of toosendanin content in toosendan seeds in Chinese Pharmacopoeia 2015 edition,a method for determination of toosendanin content in Sanzisan was established by using HPLCELSD instrument.Results:The chromatographic conditions of HPLC-ELSD were determined as follows:C18 column,acetonitrile-water(30∶70)as mobile phase,flow rate:1.0 mL/min,column temperature:45℃,detector drift tube temperature:100.0℃,gas flow rate:2.8 L/min.Methodological investigation showed that the method had good precision,accuracy and repeatability.Under the conditions of this study,the linear relationship of toosendanin was good(r^(2)=0.995)in the range of 3.42~10.70μg,the average recovery was 97.2%,RSD=1.3%(n=6).Conclusion:A method for the determination of toosendanin in Sanzisan by HPLC-ELSD was established for the first time.The method is sensitive,rapid and accurate,and suitable for the determination of toosendanin in Sanzisan.
