摘要
目的探讨结直肠癌组织和细胞中人整合素α6的表达特点及作用。方法以人结直肠癌细胞株HT-29、RKO,人永生化结肠上皮细胞系IMCE,以及结直肠癌组织和癌旁正常组织为研究对象。Western blot检测整合素α6在结直肠癌组织和癌细胞中的表达特点;使用整合素α6慢病毒过表达载体转染结直肠癌细胞,进行Transwell细胞迁移和侵袭实验;同时检测整合素α6过表达对结直肠癌细胞E-钙黏蛋白、N-钙黏蛋白、波形蛋白、p-Smad2及Smad2蛋白表达水平。结果结直肠癌HT-29细胞、RKO细胞中整合素α6表达水平明显高于人永生化结肠上皮IMCE细胞,差异有统计学意义(P<0.05);结直肠癌组织中整合素α6蛋白水平明显高于癌旁正常组织,差异有统计学意义(P<0.05)。结直肠癌细胞HT-29、RKO中整合素α6过表达组迁移细胞数和侵袭细胞数均明显高于空白对照组,差异有统计学意义(P<0.05)。结直肠癌细胞HT-29、RKO中整合素α6过表达组E-钙黏蛋白表达水平明显低于空白对照组,N-钙黏蛋白、波形蛋白表达水平明显高于空白对照组,差异有统计学意义(P<0.05)。结直肠癌细胞HT-29、RKO中整合素α6过表达组p-Smad2水平明显高于空白对照组,差异有统计学意义(P<0.05)。结论整合素α6在结直肠癌中高表达,可通过激活TGF-β/Smad2信号通路促进肿瘤细胞的上皮细胞-间充质转化和转移。
Objective To investigate the expression of human Integrinα6 in colorectal cancer tissues and cells and function analysis.Methods Human colorectal cancer cell lines HT-29 and RKO,human immortalized colorectal epithelial cell line IMCE,and colorectal cancer tissues and adjacent normal tissues were selected as research objects.The expression characteristics of Integrinα6 in colorectal cancer tissues and cancer cells by Western blot;Using Integrinsα6 lentivirus overexpression vector was transfected into colorectal cancer cells for Transwell cell migration and invasion experiment;At the same time,E-cadherin,N-cadherin,vimentin,p-Smad2 and Smad2 protein expression was detected in colorectal cancer cells overexpressing integrinα6.Results Integrinα6 was expressed in HT-29 cells and RKO cells was significantly higher than that of IMCE cells(P<0.05);In colorectal cancer tissue the level of Integrinα6 protein was significantly higher than that of normal tissues(P<0.05).Expression of Integrinα6 in HT-29 and RKO cells of colorectal cancer,the number of migrating cells and invasive cells in the overexpression group was significantly higher than that in the blank control group(P<0.05).Expression of Integrinα6 in HT-29 and RKO cells of colorectal cancer,the expression level of E-cadherin in overexpression group was significantly lower than that in blank control group,and the expression levels of N-cadherin and vimentin in overexpression group were significantly higher than that in blank control group(P<0.05).Expression of integrinα6 in HT-29 and RKO cells of colorectal cancer Integrinα6 the level of p-Smad2 in overexpression group was significantly higher than that in blank control group(P<0.05).Conclusion Integrinα6 is highly expressed in colorectal cancer and can promote the epithelial-mesenchymal transition and metastasis of tumor cells by activating the TGF-β/Smad2 signaling pathway.
