摘要
以‘中秋酥脆枣’为试材,采用体视显微镜、扫描电子显微镜以及花粉离体培养法对不同发育时期雄蕊形态发育特性、花粉形态特征和花粉活力等进行研究。探明其相互之间对应的关系及花粉的适宜贮藏条件,旨在为提高枣树杂交育种效率提供依据。结果表明‘中秋酥脆枣’花药呈椭圆形,表面有网状纹饰,2个花粉囊对称,纵向开裂;花粉属于小型花粉,外壁呈网状雕纹,近扁球形或近球形,极面观呈正三角形,赤道面观为椭圆形;萌发孔3个,萌发沟3裂,延伸至两极但无相连通。花粉萌发率与花粉的发育成熟度存在一定的关系。初开期、萼片展平期和花瓣花药分离期花粉萌发率较高。低温干燥贮藏花粉效果较好。
In order to provide basis for improving the efficiency of jujube hybrid breeding,we took flowers of Ziziphus jujuba Zhongqiusucui at different development stages as material,studied the stamen morphological development and pollen viability of‘Zhongqiusucui’,and found out their relationship and the best pollen collection time and storage conditions.Stereomicroscope and scanning electron microscopes(SEM)were used to study the characteristics of stamen development and pollen morphology at flowers different development stages.Pollen viability was measured by in vitro culture.The results showed that the pollens of‘Zhongqiusucui’were oval,with reticulate ornamentation on the surface,symmetrical pollen sacs and longitudinal dehiscence.Pollen belonged to small pollens,which was nearly flat spherical or nearly spherical,polar view was regular triangle and equatorial view was elliptic.There were three germinating holes and three germinating grooves,which extended to the poles but were not connected.The outer wall of pollen grains was reticulate,with shallow reticular,irregular and large meshes.Pollen germination rate had a certain relationship with pollen development maturity.The pollen germination rate was higher in the first blossoming stage,sepal flattening stage and petal anther separation stage.The effect of low temperature drying on storage of pollen was better.
