摘要
目的:建立蜘蛛香UPLC指纹图谱,为其质量控制提供科学依据。方法:采用Ultimate~? UHPLC Polar-RP色谱柱(100 mm×2.1 mm,1.8μm),以乙腈-0.1%甲酸溶液为流动相,梯度洗脱;检测波长为327 nm(0~11 min)和256 nm(11~30 min);流速为0.21 mL/min;柱温为30℃。通过相似度评价、聚类分析、主成分分析、偏最小二乘判别分析法对21批蜘蛛香指纹图谱进行分析。结果:建立了蜘蛛香的UPLC指纹图谱,确定了25个共有峰,指认了9个成分,其中16批样品相似度大于0.900,所有样品可分成2类,并发现缬草三酯、绿原酸、异绿原酸A、乙酰缬草三酯、异绿原酸C是样品间差异较大的5个成分。结论:UPLC指纹图谱结合化学模式识别能较好地反映蜘蛛香的整体质量,可为蜘蛛香质量控制和评价提供参考。
Objective:To establish the UPLC fingerprint of Valeriana jatamansi in order to provide a reference for its quality control.Methods:The analysis was performed on Ultimate~? UHPLC Polar-RP(100 mm×2.1 mm,1.8 μm)column with acetonitrile-0.1% formic acid solution as the mobile phase for gradient elution,the detection wavelength was 327 nm(0~11 min)and 256 nm(11~30 min),the flow rate was 0.21 mL/min,and the column temperature was 30 ℃.The fingerprints of 21 batches of Valeriana jatamansi were analyzed by similarity evaluation,hierarchical clustering analysis,principal component analysis and orthogonal partial least squares discriminant analysis.Results:The UPLC fingerprint of Valeriana jatamansi was established.There were twenty-five common peaks in the fingerprints and nine common peaks were identified.The similarity of 16 batches of samples were over 0.900,all samples were classified into two groups.Five components were the main markers that caused differences in the different batches of samples,including valepotriate,chlorogenic acid,isochlorogenic acid A,acevaltratum,isochlorogenic acid C.Conclusion:UPLC fingerprint combining with recognition of chemical pattern can reflect the intrinsic quality of Valeriana jatamansi,which may provide the reference for quality control and evaluation of Valeriana jatamansi.
