摘要
目的:建立长白瑞香HPLC指纹图谱。方法:采用SunFireTM C18(250 mm×4.6 mm,5μm)色谱柱;以乙腈-0.05%磷酸溶液为流动相,梯度洗脱;检测波长为327 nm;柱温为40℃;进样量为10μL;流速为1.0 mL/min;建立10批不同产地的长白瑞香指纹图谱,进行相似度评价,结合化学计量学方法对长白瑞香进行质量研究。结果:建立的指纹图谱共标定34个共有峰,指认其中3个成分,其相似度均在0.950以上;聚类分析可将样品聚为2类,结合主成分分析、正交偏最小二乘判别分析发现其中14个成分是造成不同批次样品差异性的主要标记物。结论:该分析方法科学、准确、简便易行,指纹图谱结合化学模式识别技术可更加系统、全面地评价长白瑞香的质量。
Objective:To establish the HPLC fingerprint of Daphne konreana.Methods:The chromatographic column was SunFireTM C18(250 mm×4.6 mm,5μm).Acetonitrile-0.05%phosphoric acid solution was used as mobile phase for gradient elution with the flow rate of 1.0 mL/min,the detection wavelength was 327 nm,column temperature was 40℃,injection volume was 10μL;The fingerprint of ten batches of Daphne konreana from different producing areas was established,the similarity was evaluated.And the chemical pattern recognition was used for quality study.Results:A total of thirty-four common peaks were demarcated in the established fingerprint,three components were identified.The similarity degree was above 0.950.Cluster analysis could cluster samples into two categories.Combined with principal component analysis,discriminant analysis of orthogonal partial least squares,it was found that fourteen components were the main markers that caused the differences of different batches of samples.Conclusion:This method is scientific,accurate,simple and easy to use.Fingerprint combined with chemical pattern recognition can systematically and comprehensively evaluate the quality of Daphne konreana.
