摘要
目的:采用网络药理学方法研究藏药复方达布班杂防治高原肺水肿(HAPE)多成分、多靶点、多途径的分子作用机制。方法:通过文献挖掘和TCMSP数据库收集达布班杂的主要化学成分,使用PharmMapper和DrugBank数据库进行靶点预测和筛选,通过DAVID和KEGG通路注释分析达布班杂治疗HAPE主要活性成分的作用靶点与通路,采用Cytoscape3.5.0软件构建达布班杂治疗HAPE"活性成分-核心靶点-通路"网络,预测其治疗HAPE关键活性成分及相关作用靶点。结果:构建的"化合物-核心靶点-通路"网络预测发现30个关键活性成分,作用于20个核心靶点,31条相关通路,主要涉及炎症反应、免疫系统、氧化应激等多种途径。推测达布班杂通过参与增强机体免疫功能、降低氧化应激损伤、恢复内皮细胞源性因子平衡状态、修复损伤的肺血管上皮细胞,达到防治HAPE的目的。结论:网络分析结果初步揭示了藏药复方达布班杂治疗HAPE多成分、多靶点、多通路的科学内涵,为进一步深入研究达布班杂药效物质基础及作用机制奠定了基础。
Objective:To investigate the molecular mechanism of multiple components,multiple targets and multiple pathways of Tibetan medicine Dabubanza in the prevention and treatment of high altitude pulmonary edema(HAPE) through network pharmacology.Methods:The main chemical compounds in Dabubanza were collected from published literatures and TCMSP database.The targets were predicted and screened by PharmMapper and DrugBank databases.DAVID and KEGG pathway annotations were used to analyze the target and pathway of major active components of Dabubanza in the treatment of HAPE.Cytoscape 3.5.0 software was used to construct "compounds-core targets-pathways" network of Dabubanza in the treatment of HAPE.The key active components and related targets for the treatment of HAPE were predicted.Results:The "compounds-core targets-pathways" network was constructed to predict 30 key active components acting on 20 core targets and 31 related pathways,mainly involving inflammatory response,immune system,oxidative stress and other pathways.It was speculated that Dabubanza could prevent and control HAPE by participating in enhancing the immune function of the body,reducing oxidative stress injury,restoring the balance of endothelial cell-derived factors and repairing the injured pulmonary vascular epithelial cells.Conclusion:The results of network analysis preliminarily reveal the scientific connotation of Dabubanza in the treatment of HAPE with multiple components,multiple targets and multiple pathways,which lay a foundation for further research on the pharmacodynamic substances and mechanism of Dabubanza.
