摘要
目的:研究阳春砂离体培养植株再生技术,建立阳春砂种苗快繁体系。方法:以阳春砂幼嫩根茎为外植体,将其横切成厚度约1 mm的薄片,放置在添加不同植物生长调节剂的MS培养基上诱导不定芽分化;将分化的芽转入增殖培养基进行增殖培养,再转入生根培养基中诱导生根,生根苗经炼苗后移栽室外。结果:阳春砂根茎薄片芽分化的数量与薄片所在的位置及培养基中的植物生长调节剂有关,距离顶端分生组织越近的薄片出芽数越多,生长调节剂CPPU比6-BA对薄片芽分化更有效,0.2 mg/L CPPU+0.1 mg/L NAA诱导产生的芽数量最多,达到3.6个;CPPU对芽增殖的效果优于6-BA,芽增殖系数最高为6.8,生根壮苗最佳培养基为附加0.2 mg/L NAA和1 g/L Ac的MS培养基;组培苗移栽成活率100%。结论:利用薄片培养技术建立的阳春砂高效植株再生体系,是阳春砂种苗快繁的新途径。
Objective:To study the in vitro culture plant regeneration technology of Amomum villosum and to establish the rapid propagation system of seedling.Methods:The young rhizomes of Amomum villosum as explants were cut into slices about 1 mm thick and placed on MS medium supplemented with different plant growth regulators to induce adventitious shoot differentiation.The differentiated shoots were transferred to the proliferation medium for proliferation culture,and then transferred to the rooting medium for induction of rooting.The rooting seedlings were transplanted outside after hardening-seedling.Results:The shoot differentiation number of thin section of Amomum villosum rhizomes was related to the location of the slices and the plant growth regulator in the medium.The closer the slices were to the apical meristem,the more shoots appeared.The growth regulator CPPU was more effective than 6-BA in shoot differentiation,and 0.2 mg/L CPPU with 0.1 mg/L NAA induced the largest number of shoots,up to 3.6.The effect of CPPU on shoot proliferation was better than that of 6-BA,and the highest shoot proliferation coefficient was 6.8.The best medium for rooting seedling was MS medium supplemented with 0.2 mg/L NAA and 1 g/L Ac.The survival rate of tissue culture seedling was 100%.Conclusion:The efficient plant regeneration system established by thin section culture technology is a new way of fast propagation of Amomum villosum seedlings.
