氮磷钾复合肥减量配施促生菌Bacillus sp.KTS-1-1对太子参块根蛋白质组的影响 被引量:1

摘要 目的:探讨氮磷钾复合肥减量配施促生菌Bacillus sp.KTS-1-1作用下太子参块根蛋白质差异表达情况,以期从组学角度初步了解其对太子参块根次生代谢的影响。方法:以太子参‘三泓1号’为研究材料进行盆栽试验,试验设置常规氮磷钾复合肥处理(TCF,含2.0 g氮磷钾复合肥的200 mL水溶液)和促生菌Bacillus sp.KTS-1-1与减施75%用量的常规氮磷钾复合肥配施处理(TCJ,本课题前期筛选的组合处理,含0.5 g氮磷钾复合肥的200 mL 3×10^(8)CFU/mL促生菌Bacillus sp.KTS-1-1菌悬液),块根采收前2个月灌根各处理液,采收后采用TMT标记结合高效液相色谱-质谱串联技术分析不同处理块根差异表达蛋白,并利用多重数据库对差异蛋白进行生物信息学分析。试剂盒(比色法)测定不同处理块根总黄酮含量。结果:TCJ/TCF共有89个蛋白表达差异显著,包括47个上调蛋白和42个下调蛋白。基于差异表达蛋白的功能富集分析共筛选到9个差异表达蛋白参与“黄酮类化合物生物合成”“苯丙烷类生物合成”“生物碱生物合成”及“环肽生物合成”过程;总黄酮含量验证结果与TMT定量蛋白质组结果相吻合。结论:氮磷钾复合肥减量配施促生菌Bacillus sp.KTS-1-1可能通过影响太子参块根次生代谢相关蛋白的表达变化进而影响其次生代谢活性成分的合成累积。 Objective:To understand the effect of the reduction of nitrogen,phosphorus and potassium compound fertilizer combined with growth-promoting bacteria Bacillus sp.KTS-1-1 on the differential proteins expressions of Pseudostellaria heterophylla root tubers,so as to preliminarily understand its effect on secondary metabolism of Pseudostellaria heterophylla root tubers.Methods:Pot experiment was carried out with Pseudostellaria heterophylla’Sanhong No.1’as the research material.The experiment set up the conventional nitrogen,phosphorus and potassium compound fertilizer treatment(TCF,200 mL aqueous solution containing 2.0 g NPK compound fertilizer)and the combination treatment of Bacillus sp.KTS-1-1 and conventional NPK compound fertilizer with 75%reduced dosage(TCJ,the combination treatment screened in the earlier stage of this project,200 mL 3×10^(8)CFU/mL Bacillus sp.KTS-1-1 containing 0.5 g NPK compound fertilizer),the root tubers were given each treatment solution by root irrigation in the first two months of harvest,the differentially expressed proteins in different root tubers were analyzed by TMT marker combined with high performance liquid chromatography-mass spectrometry tandem technique after harvest.The differentially expressed proteins were analyzed by bioinformatics using multiple databases.Kit(colorimetric method)was used to determine the content of total flavonoids in root tubers of different treatments.Results:There existed 89 differentially expressive proteins in TCJ/TCF,which included 47 up-regulated proteins and 42 down-regulated proteins.9 differentially expressive proteins involved in the biological synthesis of flavonoids,phenylpropanes,alkaloids and cyclic peptides were obtained based on functional enrichment analysis of differentially expressive proteins.The verification result of total flavonoids contents coincided with the result of TMT-based quantitative proteomics.Conclusion:The nitrogen,phosphorus and potassium compound fertilizer combined with growth-promoting bacteria Bacillus sp.KTS-1-1 may affect the synthesis and accumulation of secondary metabolites of Pseudostellaria heterophylla root tubers through interfering with the protein expressions related to secondary metabolites in root tubers.
