摘要
目的:研究藏药亚吉玛(长梗金腰)的化学成分。方法:采用ODS、MCI、Sephadex LH-20葡聚糖凝胶、制备/半制备HPLC等柱色谱技术从长梗金腰中分离化合物,根据其理化性质和NMR、MS波谱数据鉴定化合物结构;采用佛波豆蔻酸乙酯刺激的大鼠中性粒细胞(PMNs)呼吸爆发细胞模型评价所分离单体的抗炎活性。结果:从长梗金腰的乙酸乙酯部位中分离得到3个多甲氧基黄酮醇衍生物,分别鉴定为:金腰苷J大写英文字母(1)、异鼠李素-3-O-β-D-葡萄糖苷(2)、oxyayanin B(3)。化合物1~3对PMNs呼吸爆发具有不同程度的抑制作用,IC_(50)值分别为9.32±0.74μmol/L、5.67±0.40μmol/L、11.20±1.12μmol/L。结论:化合物1为新化合物,化合物2、3为首次从该植物中分离得到。化合物1~3具有作为抗炎药物的潜力。
Objective: To investigate the chemical constituents of Tibetan medicine "Yajima" Chrysosplenium axillare.Methods: The compounds from Chrysosplenium axillare were isolated by ODS,MCI,Sephadex LH-20 and preparative/semi-preparative HPLC.The structures of the compounds were identified according to the physical and chemical properties, NMR and MS spectra.The anti-inflammatory activity of the isolated monomers was evaluated by using a respiratory burst cell model of rat neutrophils(PMNs) stimulated with phorbol myristate acetate.Results: Three polymethoxy flavonol derivatives were isolated from ethyl acetate extract of Chrysosplenium axillare and identified as chrysosplenoside J(1),isorhamnetin-3-O-β-D-glucoside(2),oxyayanin B(3).Compounds 1 to 3 had different inhibitory effects on PMNs respiratory burst, with IC_(50)values of 9.32±0.74 μmol/L,5.67±0.40 μmol/L,11.20±1.12 μmol/L,respectively.Conclusion: Among them, compound 1 is a new compound, compounds 2 and 3 are isolated from Chrysosplenium axillare for the first time, compounds 1 to 3 have potential as anti-inflammatory agents.
