温胆汤对精神分裂症模型大鼠学习记忆及PI3K/Akt/mTOR信号通路的影响 被引量:6

Effect of Wendan Decoction on Learning and Memory and PI3K/Akt/mTOR Signaling Pathway in Schizophrenia Model Rats

摘要 目的:探讨温胆汤对精神分裂症模型大鼠学习记忆及PI3K/Akt/mTOR信号通路的影响。方法:将60只SPF级SD大鼠随机分为正常对照组、模型组、氯氮平组(20 mg/kg)及温胆汤低(10 g/kg)、中(20 g/kg)、高(40 g/kg)剂量组,各给药组灌胃给药,正常对照组与模型组灌胃相应体积生理盐水,1次/d,共14 d。除正常对照组外其余各组大鼠末次给药2 h后于左侧腹腔一次性注射MK-801(0.6 mg/kg)诱发精神分裂症模型,正常对照组腹腔注射相应体积生理盐水。Morris水迷宫实验检测各组大鼠学习记忆能力;Western Blot检测海马组织PI3K、Akt、mTOR蛋白表达;qRT-PCR检测海马组织PI3K、Akt、mTOR mRNA表达。结果:与模型组比较,除温胆汤低剂量组外,各给药组大鼠造模后第4天定位航行实验逃避潜伏期显著缩短,空间探索实验穿越平台位置次数、穿越平台所在象限次数及平台所在象限停留时间显著增多,各给药组大鼠海马组织p-PI3K/PI3K、p-Akt/Akt、p-mTOR/mTOR水平均显著降低,温胆汤高剂量组及氯氮平组PI3K、Akt、mTOR mRNA表达显著降低(P<0.05或P<0.01)。结论:温胆汤可能通过降低精神分裂症模型大鼠海马组织PI3K、Akt、mTOR蛋白磷酸化和PI3K、Akt、mTOR mRNA表达,从而调控PI3K/Akt/mTOR信号通路,提高学习记忆能力,治疗精神分裂症。 Objective: To investigate the effect of Wendan decoction on learning and memory and PI3K/Akt/mTOR signaling pathway in schizophrenia model rats.Methods: 60 SPF SD rats were randomly divided into normal control group, model group, clozapine group(20 mg/kg),Wendan decoction low(10 g/kg),medium(20 g/kg),high(40 g/kg) dose groups.The drug was administered by gavage in each administration group, and the corresponding volume of normal saline was administered by gavage in normal control group and model group once a day for 14 days.MK-801(0.6 mg/kg) was injected into the left intraperitoneal cavity after the last administration 2 hours to induce schizophrenia model, and the normal control group was intraperitoneally injected with corresponding volume of normal saline.Morris water maze test was used to detect the learning and memory ability of rats in each group.The proteins expressions of PI3K,Akt and mTOR in hippocampus were detected by Western Blot.The mRNA expressions of PI3K,Akt and mTOR in hippocampus were detected by qRT-PCR.Results: Compared with the model group, except for the low-dose Wendan decoction group, the escape latency of the positioning navigation experiment was significantly shortened on the 4 th day after the model establishment of the rats in each administration group, the times of crossing the platform position, crossing the quadrant of the platform and residence time of the quadrant of the platform were significantly increased.The levels of p-PI3K/PI3K,p-Akt/Akt and p-mTOR/mTOR in hippocampus of rats in each drug drug group were significantly decreased, the mRNA expressions of PI3K,Akt and mTOR in Wendan decoction high dose group and clozapine group were significantly decreased(P<0.05 or P<0.01).Conclusion: Wendan decoction may regulate the PI3K/Akt/mTOR signaling pathway, improve the learning and memory ability and treat schizophrenia by reducing the PI3K,Akt, mTOR proteins phosphorylation and expressions of PI3K,Akt, mTOR mRNA in the hippocampus of schizophrenia model rats.
