摘要
丝氨酸蛋白酶抑制剂B9(serine proteinase inhibitor B9,Serpin B9)是丝氨酸蛋白酶抑制剂超家族成员之一,广泛存在于动物、植物和微生物体内,其通过抑制淋巴细胞释放颗粒酶B的杀伤作用而保护细胞。研究表明,Serpin B9参与调节机体免疫、凝血和炎性反应等病理和生理过程,并且与痴呆、肝硬化、肺气肿以及肿瘤等疾病的发生和发展密切相关。近年来越来越多的研究认为,Serpin B9能够调控肿瘤细胞凋亡、肿瘤免疫、肿瘤发生和发展、肿瘤转移以及肿瘤耐药,并且能够维持肿瘤干细胞的干性,与肿瘤患者的不良预后明显相关。因此,Serpin B9可能成为潜在的肿瘤治疗靶点以及肿瘤诊断和预后判断的指标。本文旨在介绍Serpin B9的结构及其生物学功能,重点阐明其在肿瘤及肿瘤干细胞中的研究进展,以期更全面地了解Serpin B9促进肿瘤进展的机制及其对肿瘤诊断治疗的临床意义。
Serine proteinase inhibitor B9(Serpin B9)is a member of the serine protease inhibitor superfamily,which widely exists in animals,plants and microorganisms.It protects cells by inhibiting the killing effect of granzyme B released by lymphocytes.Studies have shown that Serpin B9 is involved in the regulation of pathological and physiological processes such as immunity,coagulation and inflammatory response,and is closely related to the occurrence and development of diseases such as dementia,liver cirrhosis,emphysema and tumor.In recent years,more and more studies have shown that Serpin B9 can regulate tumor cell apoptosis,immunity,tumor occurrence and development,tumor metastasis and tumor drug resistance,and can maintain the stemness of tumor stem cells,which is significantly related to the poor prognosis of patients with malignant tumor.Therefore,Serpin B9 may become a potential target for tumor treatment and an indicator for tumor diagnosis and prognosis evaluation.The purpose of this review is to introduce the structure and biological function of Serpin B9,and to focus on the research progress in Serpin B9 in tumor and tumor stem cells,in order to comprehensively understand the mechanism of Serpin B9 promoting tumor progression and its clinical significance in tumor diagnosis and treatment.
