摘要
目的研究淫羊藿女贞子配伍调节哮喘大鼠内源性性激素水平的作用机制。方法SD雄性大鼠,随机分为5组,分别是正常组、模型组、淫羊藿组、女贞子组、配伍组。使用卵蛋白致敏及激发建立大鼠哮喘模型,灌胃给药2周。肺组织采用苏木精—伊红、马松三色及高碘酸—希夫染色观察气道病理改变;靶向代谢组学检测尿液中性激素代谢水平;免疫组化法检测肺雌激素受体(estrogen receptor,ER)α、ERβ、雄激素受体蛋白表达。结果女贞子组能显著降低模型组升高的标准化气道总管壁厚度、标准化气道平滑肌厚度(P<0.01);各给药组均能降低胶原纤维增生、杯状细胞化生水平(P<0.01),降低模型组升高的尿液雌二醇水平及肺组织ERα的蛋白表达(P<0.01);淫羊藿组、配伍组能升高模型组尿液降低的睾酮水平(P<0.05或P<0.01);配伍组能升高模型组降低的肺组织雄激素受体的表达(P<0.01)。结论淫羊藿女贞子配伍防治哮喘的作用机制与调节性激素水平,改善肺组织性激素受体表达有关。
Objective To elucidate the mechanism of the combination of Epimedii Folium(EF)and Ligustrum lucidum Fructus(LLF)modulating endogenous sex hormone levels in asthmatic rats.Methods Male Sprague-Dawley(SD)rats were randomly divided into the normal group,the model group,the EF group,the LLF group and the EF&LLF group.The rat asthma model was induced through ovalbumin sensitization and challenge,followed by oral administration of the respective drugs for two weeks.Histopathological changes in the airways were assessed using the methods of Hematoxylin and Eosin(HE),Masson’s trichrome(Masson),and Periodic Acid-Schiff(PAS)staining.Targeted metabolomics were employed to measure sex hormone metabolic levels in urinary.Immunohistochemistry was utilized to determine the expression of lung estrogen receptor(ER)α,ERβ,and androgen receptor(AR).Results LLF could significantly mitigate the elevated standardized total airway wall thickness and standardized airway smooth muscle thickness observed in the model group(P<0.01).All administration groups effectively diminished collagen fiber proliferation and goblet cell metaplasia levels(P<0.01),and reduced the urinary estradiol levels and lung tissue ERαprotein expression compared with the model group(P<0.01).However,the testosterone levels in urine was increased in the EF group and the EF&LLF group(P<0.05,P<0.01),and the expression of AR was increased in the EF&LLF group in the lung tissue compared with the model group(P<0.01).Conclusion The preventive and therapeutic mechanisms of EF and LLF against asthma are associated with the regulation of sex hormone levels and the balance of sex hormone receptors in lung tissue.
