Kanamycin was used to screen To seeds of the variety Dongnong 46 transformed by means of pollen-tube method. The results showed that 400 mg·L^-1 kanamycin could inhibit growth of non-transgenic plants, and 2 positive plants were gotten combined with Gus dyeing and PCR detection. It is proved that this method is economic and effective in preliminary screening the transgenic plants. 展开更多

Photosynthetic Characteristics of Clerodendrum trichotomum Thumb. Responses to Drought, Salt and Water-logging Stresses 被引量:1

In order to reveal the photosynthetic characteristics of C. trichotomum responses to drought, salt and water-logging stresses, one-year-old potted seedlings were taken as materials, and the several stresses including natural drought, submergence stress, water-logging and different salt treatments (0.2%, 0.4%, 0.6% and 0.8% NaCl) were carried out on August 15, 2012. The morphological and photosynthetic characteristics were observed and determined. The results showed that adverse enviromental stress had a significant effect on the morphological changes and photosynthetic characteristics of C. trichotomum. On the 14th day after natural drought, the leaves wilted and could not recovery at night, and 60% of the seedlings could recover after re-watering. From the 7th day to the 10th day after submergence stress treatment, the 2nd and the 3rd leaves at the base of 60% seedling turned yellow and the lenticels were observed. At the early stage of water-logging stress, white lenticels appeared at the base of seedlings, and the leaves wilted, chlorina and fallen off on the 8th day. A large number of leaves fallen off under 0.6% NaCl or more salt stress, and even the whole plant died. The chlorophyll content, net photosynthetic rate (Pn) and transpiration rate (Tr) decreased gradually with the stress process, such as 8 days after natural drought, less than 0.4% salt stress and water-logging stress, but the changes were not significant compared with those of the control. With the increase of the stress intensity and the prolonged time, the changes of photosynthetic index were significant. All the results indicated that C. trichotomum had a certain degree of tolerance to drought, water and salt, but it was not suitable for living, in water-logging condition for a long time. 展开更多

Forestation in the increasingly urbanized area generates a huge demand for ornamental tree stocks, requiring new approach to promote the cultural efficiency of highly valued seedlings. Chitosan oligosaccharide(COS) is one of biodegradable natural plant growth modifier derived from chitin and an abundant water insoluble biopolymer. In this study, Buddhist pine(Podocarpus macrophyllus)(PM) and Northeast yew(Taxus cuspidata)(TC) were cultured with or without COS addition from 1 July to 17 November 2016. Relative to the TC seedlings, the PM seedlings had greater growth of height and biomass which was found to be negatively correlated with the changes of nitrogen(N) and phosphorus(P) concentrations in shoot during the culture. Only those TC seedlings treated without COS addition had higher proportion of decline in the initial N concentration than that in the PM seedlings. Root P concentration declined less in Buddhist pine seedlings than that in Northeast yew seedlings treated with COS. By 17 of November 2016, both N and P seemed to have been diluted in the PM seedlings and exceed the demand in the TC seedlings. In conclusion, COS had the potential to be used for the culture of ornamental tree seedlings to promote nutrient utilization for shoot biomass accumulation, but more specific effect on nutrient allocation and utilization need to be confirmed by more studies. 展开更多

In order to optimize the ultrasonic extraction technique for the total flavonoid of leaf yellows plus, the contents of 21 leaf yellows plus total flavonoid from four regions in Heilongjiang Province were comparatively analyzed. The ultrasonic extraction technology was optimized by Box-Behnken response surface method, and the total flavonoid content of 21 kinds of Acanthopanax senticosus(Rupr. et Maxim.) Harms from different producing areas were analyzed by cluster analysis. The optimal process conditions were determined as ultrasonic time 30 min, solid-liquid ratio 1 : 12 and ultrasonic power 250 W, and the average yield of the total flavonoid was 1.453 mg·g^ (-1). By optimizing the ultrasonic-assisted extraction method, the total flavonoid content from different producing areas was compared in the experiment, which provided certain data support for the optimization of the extraction process in the future and laid a certain theoretical foundation for the quality analysis of Chinese medicinal materials. 展开更多
