Correlations Between Polymorphisms of Extracellular Superoxide Dismutase, Aldehyde Dehydrogenase-2 Genes, as Well as Drinking Behavior and Pancreatic Cancer

Objective To investigate the correlation between drinking behavior combined with polymorphisms of extracellular superoxide dismutase (EC-SOD) and aldehyde dehydrogenase-2 (ALDH2) genes and pancreatic cancer. Methods The genetic polymorphisms of EC-SOD and ALDH2 were analyzed by polymerase chain reaction restriction fragment length polymorphism in the peripheral blood leukocytes obtained from 680 pancreatic cancer cases and 680 non-cancer controls. Subsequently the frequency of genotype was compared between the pancreatic cancer patients and the healthy controls.The relationship of drinking with pancreatic cancer was analyzed. Results The frequencies of EC-SOD (C/G) and ALDH2 variant genotypes were 37.35% and 68.82% respectively in the pancreatic cancer cases, and were significantly higher than those in the healthy controls (21.03% and 44.56%, all P〈0.01). People who carried EC-SOD (C/G) (0R=2.24, 95% C1= 1.81-4.03, P〈0.01) or ALDH2 variant genotypes (OR=2.75, 95% CI=1.92-4.47, P〈0.01) had a high risk to develop pancreatic cancer. Those who carried EC-SOD (C/G) genotype combined with ALDH2 variant genotype had a high risk for pancreatic cancer (29.56% vs. 6.76%, 0R=7.69, 95% CI=3.58-10.51, P〈0.01). The drinking rate of the pancreatic cancer group (64.12%) was significantly higher than that of the control group (40.15%; OR=2.66, 95% CI=1.30-4.42, P〈0.01). An interaction between drinking and EC-SOD (C/G)/ALDH2 variant genotypes increased the risk of occurrence of pancreatic cancer (OR=25.00, 95% CI= 11.87-35.64, P〈0.01). Conclusion EC-SOD (C/G), ALDH2 variant genotypes and drinking might be the risk factors of pancreatic cancer. 展开更多

Inhibition effect of expression of Cu/Zn superoxide dismutase from rice on synthesis of Glutathione in Saccharomyces cerevisiae

The expression of a rice Cu/Zn superoxide dismutase (Cu/Zn-SOD) in Saccharomyces cerevisiae regulated by GAPDH promoter, involved in the inhibition of endogenous Glutathione (GSH) synthesis, and the competitive expression was detected by constructing the expression vector transferred Cu/Zn-SOD gene into wild-type S. cerevisiae. Transcription and expression of the Cu/Zn-SOD gene in S. cerevisiawere were confirmed by northern blot and SDS-PAGE, respectively, and activity of the Cu/Zn-SOD from crude extracts was enzymatically detected based on the effect of nitroblue tetrazolium (NBT) after running a native polyacrylamide gel. The GSH synthesis was also tested by DTNB (5, 5'-Dithiobis (2-nitrobenzoic acid)) method. Results showed that GSH synthesis was evidently suppressed by the expression of Cu/Zn-SOD gene in both control and heat shock strains. It implied that the expression of the Cu/Zn-SOD gene in S. cerevisiae has more potential facility in response to oxidative exposure than that of endogenous GSH, although Cu/Zn-SOD and GSH were both contributed to the function of oxygen radical oxidoreduction. 展开更多

Though bacteria of the radiation-resistant Deinococcus radiodurans have a high resistance to the lethal and mutagenic effects of many DNA-damaging agents, the mechanisms involved in the response of these bacteria to oxidative stress are poorly understood. In this report, the superoxide dismutase (SOD) and catalase (CAT) activities produced by these bacteria were measured, and the change of SOD and CAT activities by 20 keV N+ beam exposure was examined. Their activities were increased by N+ beam exposure from 8x 1014 ions/cm2 to 6x1015 ions/cm2.The treatment of H2O2 and [ CHC13 +CH3 CH2OH ] and the measurement of absorption spectrum showed that the increase in SOD activity was resulted from inducible activities of MnSOD in D. radiodurans AS1.633 by N+ beam exposure . These results suggested that this bacteria possess inducible defense mechanisms against the deleterious effects of oxidization. 展开更多

Physiological characteristics changes of Aesculus chinensis seeds during natural dehydration 被引量:8

A study was conducted to determine the physiological characteristics changes of Aesculus chinensis seeds during natural dehydration in 2003. The results showed that A. chinensis seeds were recalcitrant with being highly desiccation-sensitive. The seed moisture content of fresh fruits was higher than 60%. When the seeds were naturally dried for 30 days, their moisture content declined to 30.2% and their viability was completely lost. The seed germination percentage had a small increase at the beginning of desiccation and then decreased rapidly. The relative electrical conductivity of the A. chinensis seeds increased along with a decrease in seed moisture content. However, there was an abnormal increase in relative electrical conductivity when the seed moisture content was between 53.7% and 50.9%. Superoxide dismutase (SOD) activity decreased rapidly in the period of desiccation except for an abnormality when the seed moisture content was between 53.7% and 50.9%. Malondialdehyde (MDA) content increased slowly at the early stage of desiccation and then rose rapidly after the moisture content was below 50.9%. The soluble sugar content in seeds slowly increased with the increasing period of desiccation. The seed germination percentage was at the high level when seed moisture content was in range of 47%- 60%, which suggests that this was the optimum moisture content for maintaining A. chinensis seed viability. 展开更多

Metal–organic frameworks(MOFs)have attracted significant research interest in biomimetic catalysis.However,the modulation of the activity of MOFs by precisely tuning the coordination of metal nodes is still a significant challenge.Inspired by metalloenzymes with well-defined coordination structures,a series of MOFs containing halogen-coordinated copper nodes(Cu-X MOFs,X=Cl,Br,I)are employed to elucidate their structure–activity relationship.Intriguingly,experimental and theoretical results strongly support that precisely tuning the coordination of halogen atoms directly regulates the enzyme-like activities of Cu-X MOFs by influencing the spatial configuration and electronic structure of the Cu active center.The optimal Cu–Cl MOF exhibits excellent superoxide dismutase-like activity with a specific activity one order of magnitude higher than the reported Cu-based nanozymes.More importantly,by performing enzyme-mimicking catalysis,the Cu–Cl MOF nanozyme can significantly scavenge reactive oxygen species and alleviate oxidative stress,thus effectively relieving ocular chemical burns.Mechanistically,the antioxidant and antiapoptotic properties of Cu–Cl MOF are achieved by regulating the NRF2 and JNK or P38 MAPK pathways.Our work provides a novel way to refine MOF nanozymes by directly engineering the coordination microenvironment and,more significantly,demonstrating their potential therapeutic effect in ophthalmic disease. 展开更多

Effects of Cold Acclimation on Several Enzyme Activities in Euonymus radicans ‘Emorald & Gold’and Its Relation to Semi-lethal Temperature 被引量:3

The changes in activities of superoxide dismutase (SOD), peroxidase (POD) and ATPase in the leaves of Euonymus radicans were studied when seedlings were cold-acclimated (at 4 ℃) for 1 week, 2 weeks, 3 weeks and then treated for 1 d under low temperature stress (at 5 ℃). The semi-lethal temperatures of acclimated and unacclimated seedlings were also investigated. The results indicated that the activities of the three enzymes in the leaves of the seedlings treated at 4 ℃ for 1, 2 and 3 weeks were all higher than those of unacclimated seedings (treated at 22 ℃ as controls). The activities of SOD and POD increased continuously with the prolongation of the time of cold acclimation, but stepped up to summits then down to the levels of the controls. The activities of SOD culminated at the first week, and the activities of POD at the second week. When acclimated and unacclimated seedlings were both treated at 5 ℃ for 1 d, the activities of the three enzymes in the leaves of acclimated seedlings were a little lower than those before stress, but higher than those of the controls. Moreover, the decrease rate of enzyme activities was greatly lower than that of the controls. The results showed that cold acclimation could enhance the stability of the three enzymes in the leaves of seedlings under low temperature stress; the semi-lethal temperature was 19.1 ℃ when the seedlings were treated at 4 ℃ for 3 weeks, but it was 5.4 ℃ when the seedlings were treated at 22 ℃. The decline of the semi-lethal temperature caused by the adaptive changes of enzyme activities was one of the foundations of enhancing the cold tolerance. 展开更多

Objective:To investigate the effect of neurotrophin-3 on the expressions of SOD and MDA in the injured spinal cord of rats. Methods: Totally 105 SD rats were randomly divided into 3 groups (n = 35): sham group, control group and experimental group. Animal model of acute spinal cord was inflicted with Allen's method by a thin plastic tube situated in subarachnoid space below the injury level for perfusion. Rats in experimental group received 20μl NT-3 (200 ng) from the tube at 0, 4. 8. 12. 24 h and 3. 7 d after injury, and those in control group got the equal volume of normal saline at the same time points. The animals in sham group only received opening vertebral plate and putting tube in subarachnoid space. The rats were sacrificed at 4, 8. 12. 24 h, and 3. 7, 14 d postinjury (n = 5). And the levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in blood were observed with colorimetric method. Results: The serum level of SOD reduced obviously and the level of MDA raised obviously in rats after the injury, and the activity of SOD reached the lowest on day 3 and the concentration of MDA reached peak at the 7 d. In the experimental group, the SOD level was obviously higher (P<0. 01). and MDA level was lower than the control (P<0. 01). Conclusion:NT-3 can mitigate secondary injury of spinal cord in vivo. One of mechanisms is that inhibits abnormal expression of MDA and elevates the activity of SOD. thus the injury of free radical and lipid peroxidation is attenuated. 展开更多

At different time-points, the normal and hepatic neoplasm tissues of the animal models were taken out to detect the superoxide dismutase (SOD) and malondialdehyde (MDA) respectively.The results show that the products and injurious effects of oxygen free radical (OFR) of the neoplasm tissues are more serious than those of the normal hepatic tissues. 展开更多

Wheat embryo globulin(WEG)has been proven to possess multiple biological activities,including antioxidative properties,immunomodulatory,and so on.Aged mouse model were established by subcutaneous injection of D-galactose(D-gal),and the effects of WEG on learning,memory,and antioxidant capacity in aging mice were explored through behavioural tests and antioxidant enzyme activities determination.Compared with the Model group,WEG improved the percentage of the platform quadrant,increased the number of crossing platforms,and enhanced the identification indexs.WEG also increased total antioxidant capacity(T-AOC),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)activities in the liver and brains of aging mice,and reduced malondialdehyde(MDA)content.Pathological observations indicated that WEG protected against damage to brain in D-gal-induced aging mice.These results effectively revealed that WEG not only improved the abilities of learning and memory,and the cognitive impairment,but also delayed the aging process of the D-gal-induced mice. 展开更多

The rats with hypertension were... Objective: To study the effects of Salvia Miltiorrhiza Bge (SMB) on myocardial fibrosis in hypertension in rats. Methods: Normal Wistar kyoto rats served as negative control (group A) . The rats with hypertension were divided into 2 groups: the hypertension group (group B) which served as the positive control and the SMB-treated hypertension group (group C). The effects of SMB on the systolic pressure, the myocardial content of aldosterone (ALD) and nitric oxide (NO) and the activity of superoxide dismutase (SOD) in the myocardium were detected. Results: The systolic pressure of the rats of group C showed no marked change after the administration of SMB and showed no difference from that of the rats of group B. The content of collagen Ⅰ and Ⅲ of the left ventricle, the collagen Ⅰ /collagen Ⅲ ratio and the content-of ALD were lower in group C than in group B and higher in group C than in group A. The content of NO and the activity of SOD in the myocardium were higher in group C than in group B and lower in group C than in group A. Conclusion: It was concluded that SMB was able to prevent or reverse the progress of myocardial fibrosis resulting from hypertension. The mechanism of this effect of SMB might be due to the fact that SMB can decrease the collagen synthesis and ALD content and increase collagen decomposition, NO content and SOD activity in the myocardium. 展开更多

Objective To investigate the radiosensitizing effect of nitric oxide(NO) combined with radiation on esophageal cancer cell line TE-1.Methods Methyl thiazolyl tetrazolium(MTT) assay was used to assess the effects of NO and radiation on TE-1 cells regarding inhibition of cell proliferation.Flow cytometry was used to examine the effect of NO and radiation on cell apoptosis and cycle.Reverse transcription polymerase chine reaction and Western blot were used to evaluete the effect of NO on mRNA and protein expression of manganese superoxide dismutase(MnSOD).Results NO inhibited the proliferation of TE-1 cells while significantly enhancing their radiosensitivity.The application of NO combined with radiation significantly increased the apoptosis rate and G2/M phase proportion of TE-1 cells,with substantial decreases in the MnSOD mRNA and protein expression levels.Conclusions NO reduces the MnSOD mRNA and protein expression levels by affecting TE-1 cell cycle,further inhibiting the apoptosis of esophageal cancer cells and enhancing the killing effect of radiation on esophageal cancer cells. 展开更多

Atorvastatin attenuates oxidative stress in Alzheimer's disease

Methods Subjects were divided into: normal blood lipid level group with Alzheimer's disease (A), higher blood lipid level group with Alzheimer's disease (AH), normal blood lipid level Alzheimer's disease group with atorvastatin treeatment (AT), higher blood lipid level Alzheimer's disease group with atorvastatin treeatment(AHT). Ox-LDL was measured by enzyme linked immunosorbent assay; SOD, MDA, ox-LDL, AchE, Ach and blood lipid level in AD was measured by biochemistry. Results: The serum level of MDA, Ache in AH group after atorvastatin treatment is lower ;The serum level of SOD, Ach in AH group is more increased than that of in A group; The serum level of ox-LDL in AH, A groups is lower than that of in A group; The dementia degree is lower after atorvastatin treatment. Conclusion: Atorvastatin can decrease serum level of MDA, AchE and ox-LDL, and increase that of SOD, Ach, and attenuate dementia symptom in AD, especially, with hyperlipemia. The hypothesis of atorvastatin neuroprotection is concluded that atorvastatin may restrain free radical reaction and retard oxidation in AD. 展开更多

Objective:To study the effect of reactive oxygen species of inducing apoptosis on the heptocacinoma tissues following ischemia and reperfusion and perfusion hyperoxia liquid of hepatocarcinoma. Methods: The hepatocarcinoma animal models ware established by implantation of VX2 tumor constitution mass into the left middle lobe of liver of rabbits. The animals were subjected to 60 min clamp-induced ischemia of hepatic artery distributing in the left middle lobe followed by reperfusion at 1 h, 1 d, 3 d and 7 d, respectively, and perfusion hyperoxia liquid (partial pressure of oxygen, PO2>80 kPa) at the same time with reperfusion beginning. The concentration of MDA and NO ware tested. Apoptotic changes in the hepatocarcinoma and normal hepatic tissues were observed by means of HE staining and terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL). Results:The concentration of MDA in normal hepatic tissues and hepatocarcinoma tissue increased followed ischemia and reperfusion especially for reperfusion 1 h (4.61±0.40, 3.10±0.23) and restored to normal level at reperfusion 7 d in normal hepatic tissues but still kept high concentration in the hepatocarcinoma tissue. Even though concentration of MDA in normal hepatic tissues is higher than that of before ischemia and reperfusion, no difference have been found after perfusion of hyperoxia liquid, and in the hepatocarcinoma tissue, the increasing of concentration of MDA was obvious after simply ischemia and reperfusion at reperfusion 1 d (4.25±0.45). The concentration of NO in normal hepatic tissues increased for reperfusion 3 d and 7 d(18.17±0.13, 17.45±0.23),while that of hepatocarcinoma tissue decreased at reperfusion 3 d(15.95±043). After perfusion of hyperoxia liquid, the concentration of NO in normal hepatic tissues kept increasing and that decreased in the hepatocarcinoma tissues in all time point and reached the lowest level at reperfusion 1 d(14.62±0.45).The result demonstrated the changes of concentration of NO and MDA in the hepatocarcinoma tissues ware more obvious than that of normal hepatic tissues(P<0.01). Conclusion:Perfusion of hyperoxia liquid from hepatic portal vein can intensify ischemia and reperfusion injury but less so for normal hepatic tissues. 展开更多
