Physiological studies of soybean [Glycine max(L.)Merr.]genotypes with wide differences in seed protein concentration may permit detection of important yield related processes.In order to research the law of protein accumulation and the characteristics of N accumulation and translocation,we did an experiment with three soybean cultivars which have different protein content and the similar phase in pot culture.The results showed that the laws of protein accumulation of three soybean cultivars are similar in the course of seeding;protein content descended in the early stage,and increased steadily in the middle period,then increased quickly in the later period.But the speed of protein accumulation in soybean seeds was difference in different period.In addition,high protein cultivar (Dongnong 42) and intermediate protein cultivar (Dongnong 7819) were more than those of low protein cultivar (Dongnong 434),including nitrogen contents in leaf and petiole,stem and pod,peak value of nitrogen accumulation of the whole plant,value of nitrogen translocation,its efficiency. 展开更多

Three different genotypes soybeans were adopted in this experiment under three fertilizer levels. The object of this study was to investigate protein accumulation regulation of soybean cultivars under the condition of different nutrient levels, and their effects on soybean yield and quality, and to provide theoretical evidence for breed, cultivation and agricultural production, also man-powered controllable locations. The concentration of N in the leaves declined after seedling stage, then increased again at stage of early flowering, and started to decrease up to leaf senescence, declined rapidly from seed-filling season to stage of yellow ripeness. The concentration of N in the stems and pod walls declined with growth stage. High seed protein genotypes exhibited higher N assimilating and partitioning during whole growth stages. Pod walls were media of N partitioning. Protein was accumulated mainly during the later period of reproductive growth stage up to harvest, so plant growth after stage of yellow ripeness could not be neglected. 展开更多

Dwarfing is useful to reduce plant height,when breeding high-yielding and non-lodging crops.In this study,a set of natural storage protein subunit-null dwarf mutants of soybean was reported that showed strongly reduced plant stature and deficiency in various 7S and 11S subunits,designated as snd1 mutants.Under normal growth conditions,the snd1 mutants showed a severe dwarf phenotype,with plant height of about 25 cm.Compared with wild-type DN47,the mutant snd1 exhibited no obvious morphological differences at the early stage of development.All the snd1 mutants examined had fewer nodes and shorter than normal internodes;the leaves were similar in shape to normal parents,but were dark-green at the mature stage.The flower size was similar to DN47;however,the flowering period was shorter than in the wild-type.Significant variation was noted for protein content,oil content of the seeds and size of seeds(weight of 100 seeds)among 17 snd1 dwarf lines.Genetic analysis indicated that the dwarfism of snd1 was controlled by a single recessive gene.The snd1 dwarf mutant had markedly different dynamic levels of the endogenous hormones gibberellin(GA),brassinosteroid,indole-3-acetic acid and abscisic acid,at the seedling stage.Exogenous GA3 treatment led to recovery of the plant height phenotype of the snd1 mutant;GA3 at 0.1 mm had the largest effect on enhancing plant height.Using molecular markers,snd1 gene was approximately mapped in an interval of 603 kb between markers Satt166 and Satt561 on chromosome 19.Snd1 mutant provided valuable material for hypoallergenic soybean breeding and the snd1 gene might be a novel gene related to plant height in soybean. 展开更多

Two-dimensional polyacrylamide gel electrophoresis reference ... A survey of petal-specific proteomes of soybean(Glycine max(L.) Merr[Non-italic].) was conducted comparing protein expression profiles in different petals. Two-dimensional polyacrylamide gel electrophoresis reference maps of protein extracts from standard petals(SP), lateral wings(LW), keel petals(KP), and reproductive organs(RO)(a mixture of stamen and carpel) were obtained. Protein expression in the three petal types was compared using Image Master TM 2 D platinum 6.0 software. This indicated that the proportion of homologous proteins between SP and LW was 59.27%, between SP and KP was 61.48%, and between LW and KP was 60.05%. Within a mass range of 6.5-200.0 ku and pH 4.0-7.0, approximately 590, 646, 544, and 700 protein spots were detected in SP, LW, KP, and RO, respectively. A total of 82 differentially expressed proteins were detected. Sixty-four of these detected spots were differentially expressed and showed more than 2-fold changes in abundance; of these 64 proteins, 26 showed increased expression and 38 showed decreased expression. Among these spots, single organ-specific proteins were also identified.They were ID 49(60.9 ku), ID 45(50.0 ku), and ID 46(40.5 ku) in RO, ID 98(42.0 ku) in SP, and ID 05(29.0 ku) in KP. A total of 14 protein spots from 82 differentially expressed proteins were identified with LC-MS/MS. Further protein identification was conducted using the SwissProt and NCBInr databases. The identified proteins and their putative functions were discussed further. This was the first study reporting the comparison of petal protein profiles of soybean florets using proteomics tools. 展开更多

Peptides from Alcalase-hydrolyzed soybean protein hydrolysate(SPH)may hold the potential as natural antioxidants.In addition,the effect of human gastrointestinal(GI)tract on peptide bioavailability needs to be explored.In this study,the impact of simulated GI digestion and transepithelial transport on various antioxidant properties of SPH were investigated.SPH displayed DPPH radical scavenging(IC50=4.22 mg/m L),ABTS·+radical scavenging(IC50=2.93 mg/m L),reducing power and metal ion-chelating activities(IC50=0.67 mg/m L).Furthermore,SPH significantly(P<0.05)inhibited the generation of intracellular reactive oxygen species(ROS)in Caco-2 cells.After simulated GI digestion,the antioxidant properties of SPH were enhanced,except for a decrease in ABTS·+radical scavenging activity.After transepithelial transport,the permeates maintained partial antioxidant activity and the LC-MS/MS data further identified the absorbed soybean peptides.These results suggest that SPH contains the antioxidant peptides that are potentially bioavailable and can be regarded as a promising source of functional food ingredients. 展开更多

Soybeans are known as a promising source of bioactive peptides.However,knowledge on the antioxidant behaviors of soybean protein hydrolysate(SPH)in the human intestinal epithelium is limited.In this study,SPH was prepared with Alcalase and subsequently ultrafiltered into four peptide fractions as SPH-I(<3 kDa),SPH-II(3~5 k Da),SPH-III(5~10 k Da)and SPH-IV(>10 kDa).The antioxidant properties of SPH and membrane fractions were investigated using different chemical assays and their protective effects against oxidative stress were evaluated using H2 O2-stressed human intestinal Caco-2 cells.Results showed that SPH-I exhibited the strongest 2,2-diphenyl-1-picrylhydrazyl(DPPH)radical scavenging activity(IC50=2.56 mg/m L)and reducing capacity while SPH-III had the best metal ion-chelating activity(IC50=0.29 mg/m L).Both SPH and the peptide fractions dose-dependently suppressed intracellular reactive oxygen species(ROS)accumulation induced by H2O2 in Caco-2 cells,but the strongest inhibitory effect was observed for SPH-I.Amino acid(AA)results revealed that SPH-I was rich in hydrophobic and antioxidant AAs,which could contribute to its stronger antioxidant properties.Additionally,SPH-I protected Caco-2 cells from H2O2-induced oxidative stress via inhibiting lipid peroxidation and stimulating antioxidant enzyme activities.These results suggest that SPH-I and constitutive peptides can be beneficial ingredients with antioxidant properties and protective effects against ROS-mediated intestinal injury. 展开更多

Genetic characters of 10 soybean breeds in Heilongjiang Province were studied using RAPD technique. We calculated the genetic similarity and genetic distance, made the cluster analysis, and also studied the genetic relationship among the 10 breeds. We found 93 amplification segments by RAPE) analysis, in which there were 70 polymorphism segments, 75.26% of amplification segments, using 13 random primers, 11, 12, 5, 4, 7, 8, 3, 7, 2, 5, 10, 10, 9 amplificated bands were found. There were 7.15 bands per primer in average, The genetic similarity among the breeds ranged from 0.6848 to 0.9019, and the genetic distance ranged from 0.0981 to 0.3152. Genetic relationship between 90035 and 9674 was the closest, and between Heifeng 27 and 90035 was the farthest. Protein analysis revealed that the closest genetic relationship was between 90035 and 90024, and the farthest was between 1330 and Sui 4, and between 1330 and Heinong 39. 展开更多

The fungal disease c... Soybean is one of the most important sources of edible oil and proteins in the world. However, it suffers from many kinds of fungal diseases which is a major limiting factor in soybean production. The fungal disease can be effectively controlled by breeding plant cultivars with genetic transformation. In this study, the resistance to Phytophthora sojae of five bivalent transgenic soybean lines was identified using the hypocotyls inoculation technique. The lines were the T2 of the transgenic soybean which were transformed with kidney bean chitinase gene and barley ribosome inactivating protein gene, and were positive by Southern Blot analysis. The resistance difference was studied through comparing the death percentage of transgenic soybean with the control. The results showed that four lines were more resistant to P sojae, whereas other one had no significant difference in comparison with the control. These transgenic soybean lines with enhanced resistance to P sojae will be useful in soybean resistance breeding. 展开更多

为研究大豆酶解蛋白替代豆粕对肉鸡生长性能、屠宰性能、肉品质、血清生化指标和抗氧化能力影响,试验选用1日龄体重相近、健康的爱拔益加雄性肉仔鸡336只,随机分成2组,每组7个重复,每个重复24只鸡。对照组饲喂基础日粮,试验组在基础日... 为研究大豆酶解蛋白替代豆粕对肉鸡生长性能、屠宰性能、肉品质、血清生化指标和抗氧化能力影响,试验选用1日龄体重相近、健康的爱拔益加雄性肉仔鸡336只,随机分成2组,每组7个重复,每个重复24只鸡。对照组饲喂基础日粮,试验组在基础日粮中添加5%大豆酶解蛋白替代豆粕,两组营养水平保持一致,试验期为40 d。结果显示:与对照组相比,大豆酶解蛋白组1~14日龄和1~40日龄肉鸡料重比显著升高(P<0.05);与对照组相比,大豆酶解蛋白组肉鸡屠宰率和半净膛率显著升高(P<0.05);与对照组相比,大豆酶解蛋白组14日龄肉鸡血清谷丙转氨酶和总胆固醇含量以及40日龄肉鸡尿素氮含量显著升高(P<0.05);与对照组相比,大豆酶解蛋白组40日龄肉鸡血浆总超氧化物歧化酶活性显著升高(P<0.05),谷胱甘肽过氧化物酶活性显著降低(P<0.05)。结果表明:饲粮添加5%大豆酶解蛋白替代豆粕对肉鸡出栏重无负面影响,但显著提高肉鸡料重比、屠宰率和抗氧化能力。 展开更多

野生大豆具有较高的籽粒蛋白含量,发掘和利用其蛋白含量基因对于高蛋白种质创制和品种培育具有重要价值。以绥农14/ZYD01015杂交组合衍生的RIL群体为材料,开展野生大豆ZYD01015的蛋白含量基因定位研究。RIL群体籽粒蛋白含量具有较大的... 野生大豆具有较高的籽粒蛋白含量,发掘和利用其蛋白含量基因对于高蛋白种质创制和品种培育具有重要价值。以绥农14/ZYD01015杂交组合衍生的RIL群体为材料,开展野生大豆ZYD01015的蛋白含量基因定位研究。RIL群体籽粒蛋白含量具有较大的遗传变异,且符合正态分布;对2020年和2021年RIL群体分别构建了高蛋白含量池和低蛋白含量池,采用BSA-seq(bulked segregant analysis sequencing,BSA-seq)技术进行了关联分析,在大豆20号染色体(Chr20)关联到1个区间(Chr20:3100000~33360000)。根据区间多态性Indel标记信息开发了12个分子标记,进行了蛋白含量QTL定位,鉴定出1个蛋白含量QTL位点qPRO-20,位于分子标记WS185和WS205之间,即Chr20:20071086~23798747区段,可解释14.00%(2020年)和13.26%(2021年)的遗传变异。QTL两侧分子标记WS185和WS205,其等位变异间蛋白含量均差异显著(P<0.01),可用于分子标记辅助筛选,能够从与ZYD01015相同等位基因型的材料中,筛选到蛋白含量在45%以上种质的概率达62.7%以上。 展开更多

大豆分离蛋白(Soy Protein Isolate,SPI)功能特性决定应用领域,加工过程中NaOH添加量影响SPI功能特性。文章究利用Matlab模拟,采用传统比例积分微分(proportional integral derivative,PID)和模糊自适应控制(Adaptive Fuzzy Control,AFC... 大豆分离蛋白(Soy Protein Isolate,SPI)功能特性决定应用领域,加工过程中NaOH添加量影响SPI功能特性。文章究利用Matlab模拟,采用传统比例积分微分(proportional integral derivative,PID)和模糊自适应控制(Adaptive Fuzzy Control,AFC)方法调控NaOH,中和大豆凝乳,并分析其对SPI结构和功能特性的影响。结果表明,pH值为7时,常规PID控制方法NaOH超调量为1.86%,调节时间为47.1 s;模糊自适应控制方法无超调量,调节时间为35.8 s。模糊自适应控制下,SPI傅里叶红外光谱吸收峰更稳定,紫外吸收特性更佳,二级结构向有序转变,表面疏水性较高,且溶解性、起泡性、泡沫稳定性、乳化性和乳化稳定性均优于常规PID控制方法。 展开更多

随着消费者对植物蛋白的需求不断增加,在植物蛋白市场中,大豆蛋白由于其营养价值和功能特性而被广泛应用。然而,工业生产的大豆饼粕中含有机溶剂,严重制约着大豆蛋白在绿色食品和有机食品领域中的应用。大豆的压榨方式主要为螺旋压榨法... 随着消费者对植物蛋白的需求不断增加,在植物蛋白市场中,大豆蛋白由于其营养价值和功能特性而被广泛应用。然而,工业生产的大豆饼粕中含有机溶剂,严重制约着大豆蛋白在绿色食品和有机食品领域中的应用。大豆的压榨方式主要为螺旋压榨法和液压冷榨压榨法,两种压榨方法对大豆饼粕中蛋白的品质均会产生影响。该研究基于此,对比螺旋压榨法和液压冷榨压榨法对大豆饼粕蛋白提取率和品质的影响,最终确定采用液压冷榨压榨法压榨大豆,获得的大豆饼粕中蛋白提取率和品质均更高。以液压冷榨压榨法压榨的大豆饼粕为原材料,对其蛋白和脂质的提取工艺进行研究和优化。结果表明,最佳的大豆饼粕蛋白和脂质提取工艺为pH值10、提取时间45 min和料液比例1∶20。 展开更多

试验旨在探究大豆低丰度蛋白对断奶小鼠生长性能和肠道微生物菌群的影响。选取21日龄断奶ICR雄性小鼠180只,随机分为5组,每组3个重复,每重复12只小鼠,每天分别灌胃含0(对照组)、0.5(D_(0.5 mg)组)、1.0(D_(1.0 mg)组)、2.0(D_(2.0 mg)组... 试验旨在探究大豆低丰度蛋白对断奶小鼠生长性能和肠道微生物菌群的影响。选取21日龄断奶ICR雄性小鼠180只,随机分为5组,每组3个重复,每重复12只小鼠,每天分别灌胃含0(对照组)、0.5(D_(0.5 mg)组)、1.0(D_(1.0 mg)组)、2.0(D_(2.0 mg)组)和4.0 mg(D_(4.0 mg)组)蛋白质的大豆低丰度蛋白提取物溶液200μL。预试期7 d,正式试验期28 d。结果显示,试验组小鼠末重均高于对照组,D_(2.0 mg)组末重极显著高于对照组(P<0.01),显著高于其他试验组(P<0.05);D_(2.0 mg)组平均日增重极显著高于对照组和D_(0.5 mg)组(P<0.01),平均日采食量显著高于对照组(P<0.05),体重增长率极显著高于对照组、D_(0.5 mg)组(P<0.01)。D_(2.0 mg)组肠道放线菌门和无壁菌门相对丰度显著低于对照组(P<0.05);D_(0.5 mg)组埃希氏杆菌属相对丰度显著高于对照组(P<0.05)。研究表明,灌胃大豆低丰度蛋白提取物能够调节小鼠肠道菌群组成,促进小鼠生长,添加剂量为2.0 mg/d时效果较好。 展开更多

The expression of protein and oil content of soybean seeds in worldwide was studied. The results shown that: Latitude and year's weather conditions affected protein content of soybean seeds importantly. Elevation affected oil content significantly. There was important difference in protein / oil content among different eco - geographical regions. The zone, in which there was the highest content of protein / oil, was that one with latitude from 0°to 20°59′ . And in this zone, there were different elevation regions, in which protein or oil content was the highest. For high pro-tein, it was the elevation region with 500 - 1000 m; for oil, it was 0~ 500 m. Protein / oil content was also different among different years. However, the changing range among years in Tropical and Subtropical regions was smaller than that in Temperate Regions. The relationship between protein / oil content and latitude, elevation or soil pH in the regions outside the Tropic of concerned and the Tropic of Capricorn was more significant than that in the region inside Regression Lines, i. e. Equatorial Rainforest and Tropical Grassland regions. 展开更多

试验旨在研究蛋鸡玉米-豆粕-棉粕-菜籽粕型低蛋白日粮的可行性并确定适宜的蛋白质水平及氨基酸平衡模式。选取400日龄的海兰褐蛋鸡504只,随机分为7组,每组6个重复,每个重复12只鸡。饲喂玉米-豆粕-棉粕-菜籽粕型日粮,Ⅰ组(对照组)粗蛋白... 试验旨在研究蛋鸡玉米-豆粕-棉粕-菜籽粕型低蛋白日粮的可行性并确定适宜的蛋白质水平及氨基酸平衡模式。选取400日龄的海兰褐蛋鸡504只,随机分为7组,每组6个重复,每个重复12只鸡。饲喂玉米-豆粕-棉粕-菜籽粕型日粮,Ⅰ组(对照组)粗蛋白水平为15.5%,Ⅱ、Ⅲ、Ⅳ组粗蛋白水平降至14.0%并分别平衡蛋氨酸(Met)+赖氨酸(Lys)、Met+Lys+苏氨酸(Thr)和Met+Lys+Thr+色氨酸(Trp),Ⅴ、Ⅵ、Ⅶ组粗蛋白水平降至12.5%并分别平衡Met+Lys、Met+Lys+Thr和Met+Lys+Thr+Trp。预试期7 d,正试期84 d。结果表明:①日粮粗蛋白水平显著影响产蛋率和料蛋比(P<0.05);氨基酸平衡模式显著影响产蛋率(P<0.05);粗蛋白水平和氨基酸平衡模式交互对产蛋率、料蛋比影响显著(P<0.05),与Ⅰ组相比,Ⅴ、Ⅵ组产蛋率显著降低(P<0.05),料蛋比显著增加(P<0.05);Ⅱ、Ⅲ、Ⅳ、Ⅶ组生产性能各项指标无显著差异(P<0.05)。②日粮粗蛋白水平和氨基酸平衡模式均显著影响粗蛋白表观代谢率(P<0.05),且交互作用显著(P<0.05),与Ⅰ组相比,Ⅲ、Ⅳ、Ⅶ组显著提高(P<0.05),Ⅱ、Ⅴ、Ⅵ组无显著差异(P<0.05)。③日粮粗蛋白水平显著影响血清总蛋白、白蛋白和尿酸含量(P<0.05);氨基酸平衡模式显著影响血清总蛋白含量(P<0.05);日粮粗蛋白水平和氨基酸平衡模式交互对血清总蛋白、白蛋白和尿酸含量影响显著(P<0.05),与Ⅰ组相比,Ⅴ、Ⅵ组总蛋白含量显著降低(P<0.05),Ⅵ组白蛋白含量显著降低(P<0.05),Ⅲ、Ⅳ、Ⅴ、Ⅵ、Ⅶ组尿酸含量显著降低(P<0.05)。④日粮粗蛋白水平和氨基酸平衡模式对粪中吲哚、粪臭素、氨气的含量影响显著(P<0.05),且交互作用显著(P<0.05),与Ⅰ组相比,其他6组吲哚、粪臭素及氨气的含量显著降低(P<0.05)。⑤Ⅱ、Ⅲ、Ⅳ组利润均高于Ⅰ组,且Ⅲ组经济效益最好。综合考虑各项指标,玉米-豆粕-棉粕-菜籽粕型低蛋白日粮具有可行性,平衡Met+Lys+Thr可使日粮粗蛋白水平降至14.0%(比正常水平降低1.5%),氨基酸平衡模式为Lys∶Met∶Thr∶Trp=100∶47∶71∶16。 展开更多

在当前豆粕减量替代的背景下,开发新型蛋白质饲料资源已成为畜牧业可持续发展的重要课题。黄酒糟作为一种富含蛋白质的酿酒副产物,通过微生物发酵技术可转化为优质饲料原料。本研究从黄酒糟中分别筛选了纤维素降解能力好、蛋白酶活性高... 在当前豆粕减量替代的背景下,开发新型蛋白质饲料资源已成为畜牧业可持续发展的重要课题。黄酒糟作为一种富含蛋白质的酿酒副产物,通过微生物发酵技术可转化为优质饲料原料。本研究从黄酒糟中分别筛选了纤维素降解能力好、蛋白酶活性高及产酸功能强的优良菌株B29、Yzz和L1,经鉴定分别为短小芽孢杆菌(Bacillus pumilus)、酿酒酵母(Saccharomyces cerevisiae)和屎肠球菌(Enterococcus faecium)。通过复合菌发酵,进一步探究了混菌协同发酵对黄酒糟饲料营养价值的改善效果。结果表明,菌剂组合B29+Yzz+L1明显提高黄酒糟饲料的营养价值:水分、乳酸、粗蛋白和酸溶蛋白含量分别增加了4.00百分点、51.11%、8.27%和38.52%,pH值降低了5.08%,活菌数达到9.3×10^(8)CFU·g^(-1)。试验结果可为黄酒糟资源化利用提供新途径,并对缓解蛋白饲料资源短缺、推动畜牧业绿色发展具有重要意义。 展开更多
