The results showed that the liquid-solid ratio was the most important factor in polysaccharides yield, followed the extraction temperature was the least important factor. The optimum microwave assisted extraction co by ndi extraction time, and tions for the highest polysaccharides yield from pumpkin (16.76%-4-0.38%) were obtained by using the response surface methodology with extraction time of 29 min, an extraction temperature of 79 ℃ and a liquid-solid ratio of 22 mL·g^-1. The best method of papain extraction were obtained, the ratio of papain 2%, pH at 5.5, temperature at 45℃ and extraction time of 20 min were suitable for papain extraction. The highest content of Hippohpae rhamoides polysaccharides was 44.28 mg·g^-1. The optimum process of ultrasonic extraction were obtained, namely extracted for 55 min at 480 W with the material ratio of 1:20. The highest content of Hippohpae rhamoides polysaccharides was 48.63 mg·g^-1. The results showed that the ultrasonic and papain extraction together was the best method, the content was 54.30 mg·g^-1. After the removing protein, pigment and dialysis. Two fraction were separated from the purified Hippohpae rhamoides by DEAE-cellulose chromatography, the main fraction was collected finally. The fraction was identified by Sepharose CL-4B gel filtration. Ultraviolet spectrometry, freeze-thawing analysis showed that fraction was purified. Its molecular weight was probably 109.4 ku.展开更多
OBJECTIVE Although the underlying mechanism is largely unknown,gut dysbiosis has emerged as a central initiator of obesity-related diseases including nonalcoholic fatty liver disease(NAFLD),type 2 diabetes and metabol...OBJECTIVE Although the underlying mechanism is largely unknown,gut dysbiosis has emerged as a central initiator of obesity-related diseases including nonalcoholic fatty liver disease(NAFLD),type 2 diabetes and metabolic syndrome.The emerging evidence support the use of prebiotics like herb-derived polysaccharides for treating NAFLD by modulating gut microbiome.So,our study focused on the microbiota-dependent anti-NAFLD effect and the exact mechanisms of Astragalus polysaccharides(APS)extracted from Astragalus mongholicus Bunge in high-fat diet(HFD)fed mice.METHODS Co-housing experiment was used to assess the microbiota dependent anti-NAFLD effect of APS.Then,targeted metabolomics and metagenomics were adopted for determining short-chain fatty acids(SCFAs)and bacteria that were specifically enriched by APS.Further in vitro experiment was carried out to test the capacity of SCFAs-producing of identified bacterium.Finally,the anti-NAFLD efficacy of identified bacterium was tested in HFD fed mice.RESULTS Our results first demonstrated the anti-NAFLD effect of APS in HFD fed mice and the contribution of gut microbiota.Moreover,our results indicated that SCFAs,predominantly acetic acid were elevated in APS-supplemented mice and ex vivo experiment.Metagenomics revealed that D.vulgaris from Desulfovibrio genus was not only enriched by APS,but also a potent generator of acetic acid,which showed significant anti-NAFLD effects in HFD fed mice.In addition,D.vulgaris modulated the hepatic gene expression pattern of lipids metabolism,particularly suppressed hepatic fatty acid synthase(FASN)and CD36 protein expression.CONCLUSION APS enriched D.vulgaris is effective on attenuating hepatic steatosis possibly through producing acetic acid,and modulation on hepatic lipids metabolism in mice.Further studies are warranted to explore the long-term impacts of D.vulgaris on host metabolism and the underlying mechanism.展开更多

The optimum experimental parameters, mechanism of the extraction and the effect of microwave-assisted extraction process on the structures of polysaccharides were investigated. The extract was analyzed by the modified phenol-sulfuric acid method at 490nm. The optimum experimental parameters were obtained by orthogonal experiments as follows: extraction time 15min, microwave radiation power 455W and the process ratio of materials mass to solvent volume 1∶20. The results show that compared with the conventional reflux extraction, the microwave-assisted extraction has a higher yield in shorter time, with no effect on the finally obtained polysaccharides as seen from the FT-IR spectra. The scanning electron microscopy images reveal that the mechanism of the extraction is related to the structural changes of the plant cells in different extracting conditions.展开更多
By a sequentially selective extraction procedure, polysaccharides (PS) from cyanobacterium Spirulina (Arthrospira) maxima that can be bio functionally healthy components were fractionated in four parts including the c...By a sequentially selective extraction procedure, polysaccharides (PS) from cyanobacterium Spirulina (Arthrospira) maxima that can be bio functionally healthy components were fractionated in four parts including the culture medium (PSCM), the external layers of the cell (PSEL), the cell wall (PSCW) and storage granules (PSSG). The four fractionated parts of polysaccharides were characterized by gas liquid chromatography (GLC) equipped with FID and fused silica capillary column (15 m×0.53 mm i. d.). The contents of PSCM, PSEL, PSCW and PSSG were about 2.0%, 2.6%, 10.0% and 52.0% to cell dry matter, respectively. Glucose was almost the only monosaccharide in PSSG and PSCW and most predominant in PSEL, while in PSCM, xylose, rhamnose and glucose were the main ones. Two uronic acids represented by glucuronic acid and galacturoic acid, six neutral monosaccharides including fucose, rhamnose, xylose, mannose, galactose and glucose, and two possible unidentified sugars were found in PSEL and PSCM. These data are valuable for the selective productions of high added value sugars from Spirulina .展开更多
OBJECTIVE To investigate the isolation,extraction,purification,and immunomodulatory activities in vitro and in vivo of polysaccharides from four different species of Dendrobiumherbs widely used in Singapore.METHODS To...OBJECTIVE To investigate the isolation,extraction,purification,and immunomodulatory activities in vitro and in vivo of polysaccharides from four different species of Dendrobiumherbs widely used in Singapore.METHODS To apply the classic method of water extraction and ethanol precipitation and the gel column chromatography technique,the polysaccharides from four species of Dendrobium herbs(D.moniliforme,D.crystallinum,D.primulinum,D.chrysanthum)were obtained.The polysaccharides were tested first on RAW 264.7 cell line with MTT method to test the effect of polysaccharides on macrophages.The two polysaccharides from D.moniliforme were found to promote proliferation of the cells,and were then tested on mice splenocyte primary cell culture.The same polysaccharides were also injected to female BALB/c mice at various doses for 21 dto see whether they would have affected the organs,especially the livers,spleens and thymuses.RESULTS Out of the 4 Dendrobiumspecies tested,it was found that polysaccharides from D.moniliformeto be most supportive of the proliferation of macrophages in a dose-dependent manner,with increasing effect with the increasing doses used.Polysaccharides from D.chrysanthumand D.crystallinum were also found to act similarly albeit to a lesser extent.The two D.moniliforme polysaccharides were also found to greatly enhance the proliferation of mouse splenocytes in vitro and growth of the spleen in vivo.CONCLUSION The immunomodulatory activities in vitro and in vivo of polysaccharides from four species of Dendrobium herbs were different.Their diverse chemical characters might contribute to their varied bioactivity,which should be further investigated.In addition,it was suggested that the polysaccharides from D.moniliforme could be potential immunostimulants used in the healthcare and food industry.展开更多

In recent years,studies have shown that there is a correlation between sleep disorders and cancer,but at the present stage,the research on sleep disorders and tumor related animal models is relatively insufficient.Our...In recent years,studies have shown that there is a correlation between sleep disorders and cancer,but at the present stage,the research on sleep disorders and tumor related animal models is relatively insufficient.Our research will focus on mice bearing B16F10-luc-G5 melanoma tumor with sleep fragmentation,detecting promoting effect of sleep fragmentation(SF)on the metastasis of melanoma.At the same time,we used Ganoderma lucidum poly⁃saccharides peptide(GL-pp,80 mg·kg-1),a component of traditional Chinese medicine Ganoderma lucidum,which has long enjoyed a good reputation at home and abroad,to observe its anti-tumor metastasis effects on B16F10-luc-G5 mice with SF.Then we used whole proteomics to analyze the difference proteins expressed in lung tissue and compared between groups,includes mice bearing B16F10-luc-G5,mice bearing B16F10-luc-G5 with SF and GL-pp administered mice bearing B16F10-luc-G5 with SF.With the analysis using bioinformatics,we found several key proteins,their genes name are Adcy9,ptk2,Yap1 and Lpin2,Per1 and Tim.And several important clusters,they are,immune system,platelet aggres⁃sion,energy metabolism,cell cytoskeleton,cell adhesion and circadian rhythms.Moreover,we detected the TLR4 signal pathway and macrophage differentiation to reconfirm the results of proteomics and trying to elucidate the mechanism of SF on tumor growth and metastasis and the effects of GL-pp.展开更多

Polysaccharide production from tea flower(TFPS) was carried out using supercritical fluid extraction(SFE).Response surface methodology(RSM),based on a five level,four variable small central composite design,was employ...Polysaccharide production from tea flower(TFPS) was carried out using supercritical fluid extraction(SFE).Response surface methodology(RSM),based on a five level,four variable small central composite design,was employed to obtain the best possible combination of extraction time,pressure,temperature and ethanol content of modifier for maximum production.The optimum conditions were as follows:extraction time of 170 min,pressure of45 MPa,temperature of 75 ℃,and 50% aqueous ethanol solution as modifier.Under these conditions,the experimental yield was 6.56 ± 0.37%,which was similar to the value predicted by the model.Monosaccharide composition of TFPS was fucose,rhamnose,arabinose,xylose,galactose,glucose,mannose,fructose,ribose,galacturonic acid and glucuronic acid in a molar percent of 31.69,0.21,0.49,1.29,35.82,0.97,1.63,18.34,7.88,1.06 and 0.63.Compared to other extraction methods,SFE could achieve higher yield and gain more types of monosaccharide.展开更多

Tea Polysaccharides(TPS) has been acknowledged of one of the most important components with various bioactivities in tea.The extraction of TPS from wide tea sources was the previous and determinant work before further...Tea Polysaccharides(TPS) has been acknowledged of one of the most important components with various bioactivities in tea.The extraction of TPS from wide tea sources was the previous and determinant work before further studies done.While the extraction procedures of TPS would be different due to the later utilization.In this work,the extraction conditions of TPS were previously optimized by respond surface methodology.A quadric model was obtained as Y = 3.16 + 0.31X1- 0.55X2- 0.47X3- 0.034X1X2+ 0.022X1X3- 0.041X2X3- 0.90X12-0.40X22- 0.73X32.The model was tested and proved through the ANOVA with F-value and p-value.Furthermore,the optimal condition was achieved as extraction temperature of 82℃,extraction time of 106 min and ratio of water to material of 33∶ 1(mL/g),meanwhile the highest yield of TPS showed 3.39% ± 0.08%.The crude TPS was later refined to be of purity of 81.3%.The refined TPS was incorporated with starch as to state the role of TPS in the starch retrogradation of starch-based food.The results showed that TPS-wheat starch mixture has a relatively low degree of retrogradation,which indicated that TPS has a delayed or resistant effect on wheat starch retrogradation.This work may provide a practical evidence for the promising starch-based food incorporated tea.展开更多
OBJECTIVE The aim of the present study was to evaluate the protective effects of momordica charantia polysaccharides(MCP) on depressive animal model induced by chronic social defeat stress(CSDS) and explore the underl...OBJECTIVE The aim of the present study was to evaluate the protective effects of momordica charantia polysaccharides(MCP) on depressive animal model induced by chronic social defeat stress(CSDS) and explore the underlying mechanisms.METHODS We established CSDS depressant mouse model and treated CSDS mice with MCP.Sucrose preference,forced swim test(FST) and social interaction test(SIT) were used to measure behaviors changes.We used ELISA,Q-PCR and western blot to test the levels of cytokines in the hippocampus.RESULTS The results showed that chronic administration of MCP(100,200 and 400 mg·kg^(-1)) significantly prevented depressive-like behaviors in mice as assessed by social interaction(SIT),tail suspension(TST) and sucrose preference tests(SPT).It was showed that the elevation of proinflammatory cytokines(TNF-α,IL-6,and IL-β) concentra.tions,up-regulation of JNK3,c-Jun,and P-110β protein expressions in the hippocampus of CSDS model.Moreover,reduction activity of PI3K and phosphorylation level of protein kinase B(AKT) was also observed in the hippocampus of CSDS model.All above phenomenon were reversed after MCP intervened.Further.more,the protective effects of MCP on the CSDS mice were partly inhibited by the specific phosphati.dylinositol 3-kinase(PI3K) inhibitor,LY294002.CONCLUSION The protective effects of MCP against depressive-like effects in CSDS mice might reduce neuroinflammatory and involve in attenuation of JNK3/PI3K/AKT pathway in the hippocampus.展开更多

文摘In recent years,studies have shown that there is a correlation between sleep disorders and cancer,but at the present stage,the research on sleep disorders and tumor related animal models is relatively insufficient.Our research will focus on mice bearing B16F10-luc-G5 melanoma tumor with sleep fragmentation,detecting promoting effect of sleep fragmentation(SF)on the metastasis of melanoma.At the same time,we used Ganoderma lucidum poly⁃saccharides peptide(GL-pp,80 mg·kg-1),a component of traditional Chinese medicine Ganoderma lucidum,which has long enjoyed a good reputation at home and abroad,to observe its anti-tumor metastasis effects on B16F10-luc-G5 mice with SF.Then we used whole proteomics to analyze the difference proteins expressed in lung tissue and compared between groups,includes mice bearing B16F10-luc-G5,mice bearing B16F10-luc-G5 with SF and GL-pp administered mice bearing B16F10-luc-G5 with SF.With the analysis using bioinformatics,we found several key proteins,their genes name are Adcy9,ptk2,Yap1 and Lpin2,Per1 and Tim.And several important clusters,they are,immune system,platelet aggres⁃sion,energy metabolism,cell cytoskeleton,cell adhesion and circadian rhythms.Moreover,we detected the TLR4 signal pathway and macrophage differentiation to reconfirm the results of proteomics and trying to elucidate the mechanism of SF on tumor growth and metastasis and the effects of GL-pp.
文摘Extracellular polysaccharide from Tremella aurantialba (TEP) significantly inhibited H2O2-induced haemolysis of RBCs, significantly decreased the amount of malondialdehyde (MDA) produced in mice liver homogenates as a result of autoxidation, and inhibited VitC-Fe2+-induced swelling of liver mitochondria.

文摘Polysaccharide production from tea flower(TFPS) was carried out using supercritical fluid extraction(SFE).Response surface methodology(RSM),based on a five level,four variable small central composite design,was employed to obtain the best possible combination of extraction time,pressure,temperature and ethanol content of modifier for maximum production.The optimum conditions were as follows:extraction time of 170 min,pressure of45 MPa,temperature of 75 ℃,and 50% aqueous ethanol solution as modifier.Under these conditions,the experimental yield was 6.56 ± 0.37%,which was similar to the value predicted by the model.Monosaccharide composition of TFPS was fucose,rhamnose,arabinose,xylose,galactose,glucose,mannose,fructose,ribose,galacturonic acid and glucuronic acid in a molar percent of 31.69,0.21,0.49,1.29,35.82,0.97,1.63,18.34,7.88,1.06 and 0.63.Compared to other extraction methods,SFE could achieve higher yield and gain more types of monosaccharide.

文摘The hypoglycemic effects of crude polysaccharides extracted from five medicinal fungi were evaluated using alloxan-induced diabetic mice.High(1 g/kg)and low(0.5 g/kg)doses of polysaccharides extracted from Cordyceps taii and C.memorabilis caused significant(P<0.01)decreases in mice blood sugar levels.Polysaccharides from Ganoderma lucidum and C.militaris fruit bodies both exhibited weak hypoglycemic effects at significance levels of P<0.01 and P<0.05 for high and low doses,respectively.Polysaccharide extracted from mycelium of a Phellinus sp.was significantly(P<0.05)hypoglycemic at higher dose levels but lower doses had no effect.

文摘Tea Polysaccharides(TPS) has been acknowledged of one of the most important components with various bioactivities in tea.The extraction of TPS from wide tea sources was the previous and determinant work before further studies done.While the extraction procedures of TPS would be different due to the later utilization.In this work,the extraction conditions of TPS were previously optimized by respond surface methodology.A quadric model was obtained as Y = 3.16 + 0.31X1- 0.55X2- 0.47X3- 0.034X1X2+ 0.022X1X3- 0.041X2X3- 0.90X12-0.40X22- 0.73X32.The model was tested and proved through the ANOVA with F-value and p-value.Furthermore,the optimal condition was achieved as extraction temperature of 82℃,extraction time of 106 min and ratio of water to material of 33∶ 1(mL/g),meanwhile the highest yield of TPS showed 3.39% ± 0.08%.The crude TPS was later refined to be of purity of 81.3%.The refined TPS was incorporated with starch as to state the role of TPS in the starch retrogradation of starch-based food.The results showed that TPS-wheat starch mixture has a relatively low degree of retrogradation,which indicated that TPS has a delayed or resistant effect on wheat starch retrogradation.This work may provide a practical evidence for the promising starch-based food incorporated tea.

文摘OBJECTIVE The aim of the present study was to evaluate the protective effects of momordica charantia polysaccharides(MCP) on depressive animal model induced by chronic social defeat stress(CSDS) and explore the underlying mechanisms.METHODS We established CSDS depressant mouse model and treated CSDS mice with MCP.Sucrose preference,forced swim test(FST) and social interaction test(SIT) were used to measure behaviors changes.We used ELISA,Q-PCR and western blot to test the levels of cytokines in the hippocampus.RESULTS The results showed that chronic administration of MCP(100,200 and 400 mg·kg^(-1)) significantly prevented depressive-like behaviors in mice as assessed by social interaction(SIT),tail suspension(TST) and sucrose preference tests(SPT).It was showed that the elevation of proinflammatory cytokines(TNF-α,IL-6,and IL-β) concentra.tions,up-regulation of JNK3,c-Jun,and P-110β protein expressions in the hippocampus of CSDS model.Moreover,reduction activity of PI3K and phosphorylation level of protein kinase B(AKT) was also observed in the hippocampus of CSDS model.All above phenomenon were reversed after MCP intervened.Further.more,the protective effects of MCP on the CSDS mice were partly inhibited by the specific phosphati.dylinositol 3-kinase(PI3K) inhibitor,LY294002.CONCLUSION The protective effects of MCP against depressive-like effects in CSDS mice might reduce neuroinflammatory and involve in attenuation of JNK3/PI3K/AKT pathway in the hippocampus.
