AIM:Normalizing the results of real-time quantitative reverse transcription polymerase chain reaction(RT-q PCR)is essential for the accuracy of analysis.Commonly used approaches include input nucleic acid standardizat...AIM:Normalizing the results of real-time quantitative reverse transcription polymerase chain reaction(RT-q PCR)is essential for the accuracy of analysis.Commonly used approaches include input nucleic acid standardization(ΔCt method),normalization against a single internal reference gene(ΔΔCt method),and geometric averaging of multiple reference gene abundance using statistical software.We evaluated these approaches in the liver of db/db mice,a typical model of fatty liver disease.METHODS:Seven reference genes,β-actin(ACTB),eukaryotic initiation factor(e IF)5,glyceraldehyde-3-phosphate dehydrogenase(GAPDH),hydroxymethylbilane synthase(HMBS),hypoxanthineguanine phosphoribosyltransferase(HPRT)1,polymerase(RNA)II(DNA directed)polypeptide A(Polr2A)and ribosomal protein P〈0(RPLP〈0),were evaluated using software of ge Norm and Norm Finder.Hepatic lipogenesis genes,such as thyroid hormone-responsive protein(Thrsp),stearoyl-Co A desaturase(SCD)1,sterol regulatory element-binding protein(SREBP)1c and fatty acid synthase(FAS),were used as target genes of interest.RESULTS:The expression levels of all target genes and GAPDH were significantly elevated(P〈0.05)in db/db mouse livers by theΔCt method.ACTB and HMBS were the most stable genes calculated by the software of ge Norm.Norm Finder analysis indicated that ACTB was the most stable gene,and the best combination of 2 genes was GAPDH and RPLP〈0.Normalization against a single internal reference gene of ACTB or RPLP〈0,the geometric mean of ACTB and HMBS,or GAPDH and RPLP〈0 showed similar results that the expression levels of Thrsp,SCD1 and FAS,but not SREBP1 c increased(P〈0.05)in the liver of db/db mice.CONCLUSION:TheΔCt approach ensures a meaningful and biologically significant appraisal of gene expression.Use of the software like ge Norm or Norm Finder should be integrated withΔCt method.展开更多
The surrounding rock of the soft rock roadway is seriously deformed and damaged under the superposition of mining stress and fault tectonic stress.In this paper,taking the No.232206 intake roadway in Meihuajing Coal M...The surrounding rock of the soft rock roadway is seriously deformed and damaged under the superposition of mining stress and fault tectonic stress.In this paper,taking the No.232206 intake roadway in Meihuajing Coal Mine as the engineering background,the deformation and failure law of the surrounding rock of the roadway in different fault protection pillar widths were obtained by numerical simulation method.On this basis,the mechanical model of the roadway under the action of hanging wall overburden migration and fault slip in normal faults was established,and the energy-driven mechanism of large deformation of the surrounding rock of the roadway was revealed.The ratio T of the energy applying on anchoring surrounding rock to the resistant energy of the anchored surrounding rock was defined as the criterion for the deformation of the roadway.Finally,it was calculated according to the actual working conditions on site,and the control method of“stress relief-support reinforcement”was used to support the roadway with the risk of large deformation.The on-site monitoring results show that the control effect of the surrounding rock of the roadway is obvious.展开更多

To investigate the applicability of four commonly used color difference formulas(CIELAB,CIE94,CMC(1:1),and CIEDE2000)in the printing field on 3D objects,as well as the impact of four standard light sources(D65,D50,A,a...To investigate the applicability of four commonly used color difference formulas(CIELAB,CIE94,CMC(1:1),and CIEDE2000)in the printing field on 3D objects,as well as the impact of four standard light sources(D65,D50,A,and TL84)on 3D color difference evaluations,50 glossy spheres with a diameter of 2cm based on the Sailner J4003D color printing device were created.These spheres were centered around the five recommended colors(gray,red,yellow,green,and blue)by CIE.Color difference was calculated according to the four formulas,and 111 pairs of experimental samples meeting the CIELAB gray scale color difference requirements(1.0-14.0)were selected.Ten observers,aged between 22 and 27 with normal color vision,were participated in this study,using the gray scale method from psychophysical experiments to conduct color difference evaluations under the four light sources,with repeated experiments for each observer.The results indicated that the overall effect of the D65 light source on 3D objects color difference was minimal.In contrast,D50 and A light sources had a significant impact within the small color difference range,while the TL84 light source influenced both large and small color difference considerably.Among the four color difference formulas,CIEDE2000 demonstrated the best predictive performance for color difference in 3D objects,followed by CMC(1:1),CIE94,and CIELAB.展开更多
Dynamic disturbances certainly reduce shear strength of rock joints,yet the mechanism needs deeper explanation.We investigate the shear behavior of a rough basalt joint by conducting laboratory shear experiments.Const...Dynamic disturbances certainly reduce shear strength of rock joints,yet the mechanism needs deeper explanation.We investigate the shear behavior of a rough basalt joint by conducting laboratory shear experiments.Constant and superimposed oscillating normal loads are applied at the upper block.Meanwhile,the bottom block moves at a constant shear rate.We investigate the shear behavior by:1)altering the normal load oscillation frequency with a same shear rate,2)altering the shear rate with a same normal load oscillation frequency,and 3)altering the normal load oscillation frequency and shear rate simultaneously with a constant ratio.The results show that the oscillating normal load reduces the coefficient of friction(COF).The reduce degree of COF increases with higher shear rate,decreases when increasing normal load oscillation frequency,and keeps constant if the special ratio,v/f(shear rate divided by normal oscillation frequency),is constant.Moreover,we identify a time lag between peak normal load and peak shear load.And the lagging proportion increases with higher shear rate,and decreases with larger static COF.Our results imply that a lower creep rate with a higher normal load oscillation frequency easily destabilizes the creeping fault zones.展开更多

The ma...LiNi1-xCoxO2 cathode materials for lithium ion batteries were synthesized by the co-precipitation and solid-state reaction methods with LiOH·H2O, Ni(NO3)2·6H2O and Co(NO3)2·6H2O as raw materials. The materials were characterized by XRD, SEM and electrochemical tests. The results showed that synthesized cathode materials were with layered structure similar to α-NaFeO2 and uniform morphology and nearly normal grain size distribution and better electrochemical performance when x was 0.18. The first charge and discharge capacity of the cathode material was 224.3 mAh·g-1 and 194.2 mAh·g-1, respectively. 88.5% of the first discharge capacity remained at the 20th cycle.展开更多
